Abstract
The invention includes an amount of (3aR)-1,3a,8-trimethyl-1,2,3,3a,8,8a-hexahydropyrrolo[2,3-b]indol-5-yl phenylcarbamate for administering to a subject and also a method of preventing or treating neurotoxicity or neurodegenerative processes in a subject in need thereof using the amount thereof.
Claims26
1-18. (canceled)
A method of reducing the amount of a neurotoxic aggregating protein in a human subject, comprising administering to a human subject a pharmaceutical composition consisting of from about 1 mg to less than about 200 mg of posiphen or a pharmaceutically acceptable salt thereof together with one or more pharmaceutically acceptable excipients, on a once a day basis.
The method of claim 16, wherein said neurotoxic aggregating protein is selected from the group consisting of APP, Aβ, SOD, Tau, alpha-synuclein (SNCA), NAC, TSE prion, and HTT.

34-37. (canceled)
39-41. (canceled)
The method of claim 41, wherein said subject is suffering from a dementia selected from the group consisting of Alzheimer's disease, Down Syndrome, alpha-synucleopathies, Parkinson's disease, Huntington's disease, Prion's disease, Amyloid Lateral Sclerosis and a tauopathy.
43-61. (canceled)
The method of claim 1, wherein the half-life of posiphen in cerebrospinal fluid after administering is about 12 hours.
The method of claim 1, wherein the half-life of posiphen in plasma after administering is about 5 hours.
The method of claim 1, wherein the pharmaceutical composition includes from about 7.5 mg to less than about 200 mg of posiphen or a pharmaceutically acceptable salt thereof.
A method of ameliorating neurodegenerative processes in a human patient, comprising administering a pharmaceutical composition consisting of from about 7.5 mg to less than about 200 mg of posiphen or a pharmaceutically acceptable salt thereof together with one or more pharmaceutically acceptable excipients, on a once a day basis, such that the production of neurotoxic aggregating protein in the human patient is inhibited.
A method of inhibiting neural alpha-synuclein expression in a human patient, comprising administering a pharmaceutical composition consisting of from about 7.5 mg to less than about 160 mg of posiphen or a pharmaceutically acceptable salt thereof together with one or more pharmaceutically acceptable excipients, the inhibition of neural alpha-synuclein expression by the posiphen being potentiated by cellular iron.
