Abstract
The present invention relates to methods for improving biophysical properties, including the stability of antibody lambda light chains, to antibody lambda light chains with improved biophysical properties, including stability, nucleic acid and vectors encoding such antibody lambda light chains, and to uses of such antibody lambda light chains, nucleic acid and vectors.
Claims10
A method for modifying the constant region of a parental antibody lambda light chain, comprising at least one of the following steps:
- (a) deleting the C-terminal amino acid residue Serine;
- (b) in the case of a lambda light chain with a constant domain Clambda2, making the following amino acid residue exchange:
- S152G, and optionally one or more of the following amino acid residue exchanges:
- A157V, T163K, Q195R, and T212A;
- (c) in the case of a lambda light chain with a constant domain Clambda7, making the following amino acid residue exchange:
- V1361, and optionally one or more of the following amino acid residue exchanges:
- T145K, K156N, V157T, and K163T.
A constant region of an antibody lambda light chain, wherein said constant region is selected from the group of:
- (i) a constant region of an antibody lambda light chain, wherein the C-terminus is amino acid residue 214;
- (ii) a constant region of an antibody lambda light chain of the type Clambda2, wherein said constant region has a glycine residue at position 152 and at least one of the following amino acid residues:
- 157V, 163K, 195R, and 212A;
- (iii) a constant region of an antibody lambda light chain of the type Clambda7, wherein said constant region has an isoleucine residue at position 136 and optionally one or more of the following amino acid residues:
- 145K, 156N, 157T, and 163T;
- (iv) a constant region exhibiting the features of both (i) and (ii);
- and
- (v) a constant region exhibiting the features of both (i) and (iii).
