Abstract
The present invention generally relates to processes for the enzymatic production of a phosphinothricin product or precursor thereof from a nitrile-containing substrate.
Claims15
An isolated nucleic acid molecule encoding an enzyme capable of catalyzing the hydrolysis of —CN to —COX, wherein X is —OH or —NH2, wherein the molecule comprises a nucleotide sequence selected from the group consisting of SEQ ID NO:
- 1, SEQ ID NO:
- 2, SEQ ID NO:
- 3, SEQ ID NO:
- 4, SEQ ID NO:
- 5, and SEQ ID NO:
- 6.
A process for the preparation of a phosphinothricin product or precursor thereof, the process comprising:
- (a) reacting acrolein with a compound of Formula II,
- thereby forming a compound of Formula III;
- (b) reacting the compound of Formula III with a cyanide source and an ammonia source, thereby forming a nitrile-containing substrate of Formula IV
- wherein R3 is hydrogen, substituted or unsubstituted C1-C8 alkyl, substituted or unsubstituted aryl, or an agronomically acceptable salt-forming cation;
- and
- (c) contacting in a reaction mixture the nitrile-containing substrate of Formula IV with an enzyme capable of catalyzing the hydrolysis of a —CN to —COX, wherein X is either —OH or —NH2, thereby forming the phosphinothricin product or precursor thereof.
A compound having the structure of Formula VII
- wherein X is either —OH or —NH2 and R3 is hydrogen, substituted or unsubstituted C1-C8 alkyl, substituted or unsubstituted aryl, or an agronomically acceptable salt-forming cation.
