Abstract
The invention provides to PKN1 gene fusions, PKN1 fusion proteins, and fragments of those genes and polypeptides. The invention further provides methods of diagnosing and treating diseases or disorders associated with PKN1 fusions, such as conditions mediated by aberrant PKN1 expression or activity, or over expression of PKN1.

1-68. (canceled)
A method for detecting in a patient a PKN1 fusion protein or a PKN1 gene fusion that results in aberrant activity or expression of PKN1 or overexpression of PKN1, wherein the PKN1 fusion protein or gene fusion is a TECR:PKN1 or ANXA4:PKN1 fusion, said method comprising:
- a) contacting a biological sample from the patient with a reagent selected from:
- i) an antibody that specifically binds to the PKN1 fusion protein, but does not specifically bind to wild-type PKN1, TECR, or ANXA4;
- or
- ii) an oligonucleotide that hybridizes to the fusion junction of the PKN1 gene fusion;
- and
- b) detecting binding between the PKN1 fusion protein or the PKN1 gene fusion and the reagent.
A method of treating a patient in which a PKN1 fusion protein or a PKN1 gene fusion selected from TECR:PKN1 and ANXA4:PKN1 has been detected, said method comprising administering to the patient a therapeutically effective amount of a compound that inhibits PKN1 activity or the activity or expression of the PKN1 fusion, wherein the PKN1 fusion has been detected in the patient by a method comprising:
- a) contacting a biological sample from the patient with a reagent selected from:
- i) an antibody that specifically binds to a PKN1 fusion protein, but does not specifically bind to wild-type PKN1, TECR, or ANXA4;
- or
- ii) an oligonucleotide that hybridizes to the fusion junction of a PKN1 gene fusion;
- and
- b) detecting binding between the PKN1 fusion protein or the PKN1 gene fusion and the reagent.
A molecule capable of specifically binding to a PKN1 fusion protein or a PKN1 gene fusion that results in aberrant activity or expression of PKN1 or overexpression PKN1, wherein the molecule is selected from:
- a) an antibody that specifically binds to the PKN1 fusion protein selected from TECR:PKN1 and ANXA4:PKN1, but does not specifically bind to wild-type PKN1, TECR, or ANXA4;
- or
- b) an oligonucleotide that hybridizes to the fusion junction of the PKN1 gene fusion selected from TECR:PKN1 and ANXA4:PKN1.
