Abstract
Described is an adenovirus derived helper virus which may comprise the adenoviral DNA sequences for E2a, S4 (orf6), the VA1 RNA gene, and the parvovirus VP capsid gene unit. Also described is a method of efficiently preparing a recombinant parvovirus (particle) which is based on the use of various adenoviral derived helper viruses/vectors.
Claims20
Adenovirus derived helper virus comprising the following adenoviral DNA sequences:
- (a) an E2a gene;
- (b) an E4(orf6) gene;
- (c) an VAI RNA gene;
- and
- (d) the parvovirus VP capsid gene unit.
2 more elements
A method of preparing a recombinant parvovirus characterized in that
- (i) mammalian cells are transfected with (a) a recombinant parvovirus vector lacking the VP1 and VP2 genes, (b) a vector containing the genes encoding the parvovirus capsid proteins, and (c) an adenovirus derived helper vector comprising the adenoviral DNA sequences from the E2a gene, the E4(orf6) gene and an VAI RNA gene;
- and
- (ii) the recombinant parvovirus is isolated from the mammalian cells or the medium after culturing the cells.
