Claims62
A ghost cell trainer (GHT), wherein said GHT comprises a plasma membrane having:
- a. a plurality of MEW molecules;
- and
- b. a plurality of a protein of interest, wherein the protein is glycosylphosphatidylinositol(GPI) anchored on the cell surface.
A ghost cell (GHC), wherein said cell comprising the following characteristics:
- a. is substantially free of all intracellular components;
- b. has a plasma membrane having:
- i. a plurality of MHC molecules;
- ii. a plurality of a protein of interest, wherein the protein is glycosylphosphatidylinositol(GPI) anchored on the cell surface.
1 more element
A method of producing a ghost cell (GHC) comprising:
- a. transfecting a cell expressing an MHC molecule with one or more plasmids comprising a gene encoding a protein of interest and a glycosylphosphatidylinositol(GPI) signaling anchor such that the protein of interest is expressed and GPI anchored to the cell surface;
- b. lysing the cells of step (a) to form a cell that is substantially free of all intracellular components.
A method of producing a ghost cell (GHC) comprising:
- a. transfecting a cell with
- i. a plasmid encoding an MHC protein and a glycosylphosphatidylinositol(GPI) signaling anchor;
- and
- ii. one or more plasmids comprising a gene encoding a protein of interest and a glycosylphosphatidylinositol(GPI) signaling anchor such that the HLA protein and the protein of interest are expressed and GPI anchored to the cell surface;
- b. lysing the cells of step (a) to form a cell that is substantially free of all intracellular components.
