Abstract
Isolated and/or purified polypeptides and nucleic acid sequences encoding polypeptides from Alicyclobacillus acidocaldarius are provided. Further provided are methods for glycosylating and/or post-translationally modifying proteins using isolated and/or purified polypeptides and nucleic acid sequences from Alicyclobacillus acidocaldarius.
Claims15
An isolated or purified polypeptide comprising a polypeptide selected from the group consisting of polypeptide having at least 90% sequence identity to SEQ ID NOS:1, 18, 35, 52, 69, 86, 103, 120, 137, 154, 171, 188, 205, 222, 239, 256, 273, 290, and 309.
A method of glycosylating or post-translationally modifying a first polypeptide at temperatures at or above about 35 Celsius or at a pH at or below about 8, the method comprising:
- providing a recombinant, purified, or isolated polypeptide encoded for by a nucleotide sequence comprising a nucleotide sequence selected from the group consisting of a nucleotide sequences having at least 90% sequence identity to at least one of the sequences of SEQ ID NOS:2, 19, 36, 53, 70, 87, 104, 121, 138, 155, 172, 189, 206, 223, 240, 257, 291, and 310;
- glycosylating or post-translationally modifying the first polypeptide with the recombinant, purified, or isolated polypeptide.
A method of modulating the protein stability, solubility, degradation, activity profile, or externalization of a first polypeptide, the methods comprising:
- providing a recombinant, purified, or isolated polypeptide encoded for by a nucleotide sequence comprising a nucleotide sequence selected from the group consisting of a nucleotide sequences having at least 90% sequence identity to at least one of the sequences of SEQ ID NOS:2, 19, 36, 53, 70, 87, 104, 121, 138, 155, 172, 189, 206, 223, 240, 257, 274, 291, and 310;
- glycosylating or post-translationally modifying the first polypeptide with the recombinant, purified, or isolated polypeptide.
