Abstract
This invention is directed to compounds, which are useful as protein kinase (PK) inhibitors and can be used to treat such diseases as cancer, blood vessel proliferative disorders, fibrotic disorders, mesangial cell proliferative disorders, metabolic diseases inflammatory disorders and neurodegenerative disorders.

A compound represented by Formula I, its enantiomers, diastereoisomers, or a pharmaceutically acceptable salt thereof:
- wherein:
- W is S;
- R1 is hydrogen, C1-8 alkyl, halo or haloalkyl;
- R2 is —N(R4)C(O)N(R4R5), —N(R4)C(O)R5, —C(O)N(R4R5), hydrogen, C1-8 alkyl, halo or haloalkyl;
- R3 is hydrogen, C1-8 alkyl, halo or haloalkyl;
- X is —N(R4)C(O)N(R4R5), —N(R4)C(O)R5, or —C(O)N(R4R5);
- R4 is hydrogen or alkyl;
- R5 is phenyl, optionally substituted with one or more substituents selected from the group consisting of methyl, ethyl, fluoro, chloro, and trifluoromethyl;
- Y is hydrogen, —C(O)—N═S(O)R7R6, —N(R4)C(O)R8, —COOR9, —C(O)NHR10, —B(OH)2, —B(OR12)(OR13) or
- R7 is C1-8 alkyl optionally substituted with one or more substituents selected from the group consisting of hydroxyl and —COOCH3, or R7 is aryl;
- R6 is C1-8 alkyl optionally substituted with one or more substituents selected from the group consisting of hydroxyl and —COOCH3, or R6 is aryl;
- R8 is C1-8 alkyl optionally substituted with one or more substituents selected from the group consisting of —COOCH3, or R8 is aryl;
- R9 is hydrogen, C1-8 alkyl, or R9 is aryl;
- R10 is hydrogen, C1-8 alkyl optionally substituted with one or more substituents selected from the group consisting of —COOCH3, or R10 is aryl;
- Z is —NHR11;
- R11 is hydrogen or C1-8 alkyl;
- R12 is hydrogen or C1-8 alkyl;
- and
- R13 is hydrogen or C1-8 alkyl.

- N-[dimethyl(oxido)-λ4-sulfanylidene]-5-{5-[({[2-fluoro-5-(trifluoromethyl)phenyl]amino}carbonyl)amino]-1-benzothien-2-yl}nicotinamide;
- and
- 5-{5-[({[4-chloro-3-(trifluoromethyl)phenyl]amino}carbonyl)amino]-1-benzothien-2-yl}-N-[dimethyl(oxido)-λ4-sulfanylidene]nicotinamide.

- R17 is hydrogen, substituted or unsubstituted C1-8 alkyl, halo or haloalkyl;
- Xa is —N(R19)C(O)N(R19R20);
- R18 is hydrogen, substituted or unsubstituted C1-8 alkyl, halo or haloalkyl;
- R19 is hydrogen, substituted or unsubstituted C1-8 alkyl, substituted or unsubstituted heterocycle or substituted or unsubstituted aryl;
- R20 is substituted or unsubstituted alkyl, substituted or unsubstituted heterocycle or substituted or unsubstituted aryl;
