Abstract
Assay supports, such as microarrays and similar devices, and methods of use and manufacture thereof, are described. The assay support is capable of assaying binding and/or activity of a bioactive entity, such as a bioactive molecule or a cell. Analytical and diagnostic uses of the supports are also described.
Claims12
An assay support for assaying selective binding or response of a cell, said support comprising:
- (a) a substrate having a surface;
- (b) one or more first layers of a polymeric material covalently attached to one or more areas of said surface, said first layers exhibiting low-cell binding properties and wherein the polymeric material is selected from the group consisting of a poly(ethylene glycol) (PEG)-based polymer, a carboxy methyl dextran, and a poly(acrylic acid);
- and
- (c) bioactive molecules covalently attached directly to one or more discrete regions of said first layers, wherein said bioactive molecules are proteins, peptides or fragments thereof capable of cell binding and wherein said bioactive molecules are covalently attached to the one or more discrete regions of said first layers in an aqueous solution.
