Abstract
Novel MSC stem-cell culture and therapy methods and culture medium compositions for the purpose of inducing, activating, or priming discrete uniform cell phenotypes to selectively promote or suppress inflammation and immunity, yielding polarized, primed, activated, or induced cells used in cell-based therapy.
Claims20
An induction media for creating an immunologically polarized mesenchymal stem cell population from an unstimulated mesenchymal stem cell population, the induction media comprising:
- a. a Toll-like receptor 3 (TLR3) ligand,
- b. erythropoietin, and
- c. hypoxia or a hypoxia mimetic,
- wherein the immunologically polarized mesenchymal stem cell population possess anti-inflammatory characteristics marked by expression of anti-inflammatory or immunosuppressive mediators.
An induction medium for creating an immunologically polarized mesenchymal stem cell population with anti-tumor properties from an unstimulated mesenchymal stem cell population, the induction medium comprising:
- a. a Toll-like receptor 4 (TLR4) ligand,
- b. erythropoietin, and
- c. 0.5-2.0% oxygen or a hypoxia mimetic, and
- d. at least one mesenchymal stem cell,
- wherein the immunologically polarized mesenchymal stem cell population possess anti-tumor pro-inflammatory characteristics marked by expression of pro-inflammatory mediators.
