Abstract
The present invention relates to various polypeptides from fish hatching fluid, their encoding nucleic acid sequences, pharmaceutical compositions comprising said polypeptides and nucleic acid molecules and their use in various medical and cosmetic applications to the skin, particularly for moisturizing skin and/or for exfoliation of the horny layer of the skin for treating or preventing skin disorders or conditions in an animal.

A cosmetic or non-cosmetic method of treating a condition or disorder of the skin of an animal wherein said skin is abnormally dry, the horny layer of the skin is abnormally thickened or the skin has a pigmentation disorder wherein a polypeptide or pharmaceutical or cosmetic composition is topically administered to said animal and wherein:
- (a) said polypeptide comprises:
- (i) an isolated polypeptide having metalloproteinase activity comprising an amino acid sequence as set forth in SEQ ID NO:
- 1;
- or
- (ii) an isolated polypeptide having metalloproteinase activity comprising an amino acid sequence which is at least 90% identical to a sequence as set forth in SEQ ID NO:
- 1;
- or
- (iii) an isolated polypeptide having metalloproteinase activity comprising a portion of an amino acid sequence as set forth in SEQ ID NO:
- 1, wherein said portion comprises at least 150 amino acids;
- or
- (iv) an isolated polypeptide having metalloproteinase activity comprising a portion of an amino acid sequence which is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 1 and comprises at least 150 amino acids;
- and
- (b) said pharmaceutical or cosmetic composition comprises a polypeptide as set forth in (a) and one or more pharmaceutically or cosmetically acceptable excipients and/or diluents.
A pharmaceutical or cosmetic composition comprising:
- (i) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 2 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 2;
- (ii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 3 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 3;
- and/or
- (iii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 4 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 4;
- (iv) one or more isolated nucleic acid molecules encoding a polypeptide as set forth in any of (i) to (iii) above or a sequence fully complementary thereof,
- an effective amount of an added stabilizing agent wherein the stabilizing agent stabilizes the isolated acidic polypeptide or the one or more isolated nucleic acid molecules from degradation,
- and one or more pharmaceutically or cosmetically acceptable excipients and/or diluents.
A cosmetic or non-cosmetic method of exfoliating and/or moisturizing skin of an animal, wherein a polypeptide or pharmaceutical or cosmetic composition is topically administered to said animal, wherein:
- (a) said polypeptide comprises:
- (i) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 2 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 2;
- (ii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 3 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 3;
- or
- (iii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 4 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 4;
- and
- (b) said pharmaceutical or cosmetic composition comprises one or more polypeptides as set forth in (a) and one or more pharmaceutically or cosmetically acceptable excipients and/or diluents.
A cosmetic or non-cosmetic method of treating a condition or disorder of the skin of an animal wherein said skin is abnormally dry, the horny layer of the skin is abnormally thickened or the skin has a pigmentation disorder wherein a polypeptide or pharmaceutical or cosmetic composition is topically administered to said animal and wherein:
- (a) said polypeptide comprises:
- (i) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 2 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 2;
- (ii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 3 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 3;
- or
- (iii) an isolated acidic polypeptide comprising an amino acid sequence as set forth in SEQ ID NO:
- 4 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion comprises at least 30 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 4;
- and
- (b) said pharmaceutical or cosmetic composition comprises one or more polypeptides as set forth in (a) and one or more pharmaceutically or cosmetically acceptable excipients and/or diluents.
An isolated acidic polypeptide consisting of:
- (i) an amino acid sequence as set forth in:
- (a) SEQ ID NO:
- 2 or 4 or a sequence which is at least 90% identical to said sequence, or a portion of any of said sequences, wherein said portion consists of at least 50 amino acids and is at least 90% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 2 or 4;
- or
- (b) SEQ ID NO:
- 3 or a sequence which is at least 99% identical to said sequence, or a portion of any of said sequences, wherein said portion consists of at least 150 amino acids and is at least 99% identical to a comparable region of an amino acid sequence as set forth in SEQ ID NO:
- 3;
- and optionally
- (ii) a flanking amino acid sequence at the N and/or C terminal of the amino acid sequence in (i) which is from 1 to 50 amino acids in length.
