Abstract
Embodiments of the present invention relate to methods for the biosynthesis of di- or trifunctional C7 alkanes in the presence of isolated enzymes or in the presence of a recombinant host cell expressing those enzymes. The di- or trifunctional C7 alkanes are useful as intermediates in the production of nylon-7, nylon-7,x, nylon-x,7, and polyesters.

A method of converting a compound, the method comprising contacting pimeloyl-CoA(PCoA) or pimeloyl [acp] (PACP) with an enzyme that catalyzes the thioester hydrolysis of PCoA or PACP to pimelic acid (PA), wherein PA is produced,
- wherein the enzyme is a thioesterase, an acid-thiol ligase, or a CoA transferase;
- and
- wherein the thioesterase is one or more of:
- the gene product of tesB or acot13;
- and
- a thioesterase in EC 3.1.2.14;
- wherein the acid-thiol ligase is an acyl-[acp}-synthetase from EC 6.2.1.14 (such as BioW) or E.C. 6.2.1.20, and
- wherein the CoA transferase is one or more of
- a CoA transferase in EC 2.8.3.14;
- and
- the gene product of thnH.
A method of converting a compound, the method comprising contacting PCoA or PACP with an enzyme that catalyzes the reduction of PCoA or PACP to PAS, wherein PAS is produced, wherein the enzyme that catalyzes the reduction of PCoA or PACP to PAS is a fatty-acyl-[acp]-reductase or an aldehyde dehyrogenase in EC 1.2.1.3, EC 1.2.1.20, EC 1.2.1.22, EC 1.2.1.57, or EC 1.2.1.76.
A method of converting a compound, the method comprising contacting α-ketosuberate (AKS) with an enzyme that catalyzes the α-keto decarboxylation of AKS to PAS, wherein PAS is produced, wherein the enzyme is an acetolactate synthase.
A method of converting a compound, the method comprising contacting AKS with an enzyme that catalyzes the transfer of an amino group to the AKS to produce α-amino suberate (AAS), wherein AAS is produced, wherein the enzyme is an aminotransferase in EC 2.6.1.21
A method of converting a compound, the method comprising contacting 7 AHT with an enzyme that catalyzes the transfer of an amino group to the 7 AHT to produce 1,7-diaminoheptane (1,7 DAH), wherein 1,7 DAH is produced, wherein the enzyme that catalyzes the transfer of an amino group to the 7 AHT to produce 1,7-diaminoheptane (1,7 DAH) is one or more of
- an amino acid aminotransferase in EC 2.6.1.21;
- a deaminating dehydrogenase;
- and
- an ω-transaminase in EC 2.6.1.11, EC 2.6.1.13, EC 2.6.1.48 or EC 2.6.1.62.
An isolated cell comprising one or more exogenous nucleic acid encoding:
- (a) (i) a thiosterase, an acid thiol ligase, or a CoA transferase;
- and (ii) a NAD-dependent non-acylating aldehyde dehyrogenase;
- (b) (i) a fatty-acyl-[acp] reductase;

- and (ii) and enzyme that catalyzes a reaction whose product is AKS;
- (d) (i) an aminotransferase;
- and (ii) an acetolactate synthase;
- or
- (e) (i) a deaminating dehydrogenase;
- and (ii) an enzyme that catalyzes a reaction whose product is 7 AHT.
