Abstract
Gene targeting is a valuable tool for basic researchers and gene therapists. Unfortunately, current methods utilised to target genes are inefficient because of their low targeting frequencies. Provided herein are methods and compositions by which gene targeting frequencies can be increased.
Claims10
A method to increase gene targeting frequency comprising inhibiting expression of at least one gene of a mismatch repair pathway or by inhibiting activity of at least one protein of a mismatch repair pathway so as to provide increased gene targeting frequency as compared to a cell in which expression and/or activity has not be inhibited.
A method to increase gene targeting frequency comprising increasing expression of at least one gene coding for Rad52, Rad57, Rad59, MUS81, XRCC3 or a combination thereof so as to provide increased gene targeting frequency as compared to a cell in which expression has not been increased.
