Abstract
Substantially human antisera are provided by genetically modifying a domestic animal generally weighing at least about 1 kg. The domestic animal is genetically modified by generating inactive heavy and light chain immunoglobulin loci and integrating at least functional portions of the human heavy and light chain immunoglobulin loci, whereby the human loci generate an immune response. The antisera find use in the treatment of diseases, immunocompromised patients and in case of transplantation.

A method of producing a substantially human polyclonal antisera composition in a transgenic rabbit, comprising:
- (a) producing a first mutated rabbit comprising heavy chain immunoglobulin loci where constant and/or variable region elements are replaced with at least a functional portion of the human heavy chain immunoglobulin locus by genetic alteration of a cell nucleus of said rabbit, introducing said cell nucleus into an enucleated nuclear transfer unit cell to provide a first embryonic stem cell, introducing said first nuclear transfer unit cell into a female rabbit host to produce a first mutated neonate;
- (b) producing a second mutated rabbit comprising light chain immunoglobulin loci where constant and/or variable region elements are replaced with at least a functional portion of the human light chain immunoglobulin locus by genetic alteration of a cell nucleus of said rabbit, introducing said cell nucleus into an enucleated nuclear transfer unit cell to provide a second embryonic cell stem cell, introducing said second nuclear transfer unit cell into a female rabbit recipient host to produce a second mutated neonate;
- (c) breeding mature first and second mutated neonates and selecting a transgenic rabbit capable of producing substantially human antisera and being substantially incapable of producing endogenous antisera;
- and
- (d) immunizing said trangenic rabbit with an immunogen to produce a substantially human polyclonal antisera composition.
A method of producing a substantially human polyclonal antisera composition in a transgenic rabbit, comprising:
- (a) producing a mutated rabbit comprising heavy and light chain immunoglobulin loci where constant and/or variable region elements are replaced with at least a functional portion or the human heavy and/or light chain immunoglobulin locus by genetic alteration of a cell nucleus of said rabbit, introducing said cell nucleus into an enucleated nuclear transfer unit cell to provide a embryonic cell stem cell, introducing said nuclear transfer unit cell into a female recipient rabbit host to produce mutated neonate;
- (b) breeding mature mutated neonates and selecting a transgenic rabbit capable of producing substantially human antisera and substantially incapable of producing endogenous antisera;
