Abstract
Methods of alleviating the symptoms of hemoglobinopathies, including, but not limited to, sickle cell disease, β-thalassemia, and hemoglobin H disease are provided. In some embodiments, the methods comprise administering an inhibitor selected from a β-arrestin1/2 inhibitor and/or a GRK2 inhibitor to the subject. Methods of inhibiting adhesion of sickle red blood cells to endothelial cells and adhesion to and activation of leukocytes are also provided.
Claims21
A method of alleviating at least one symptom of a hemoglobinopathy in a subject comprising administering an inhibitor selected from a β-arrestin1/2 inhibitor and a GRK2 inhibitor to the subject.
A method of inhibiting adhesion of cells comprising administering an inhibitor selected from a β-arrestin1/2 inhibitor and a GRK2 inhibitor to a subject to inhibit adhesion of cells.
A method of alleviating at least one of acute or chronic pain, chronic hemolysis (aplastic crises), avascular necrosis, organ damage, and erythroid hyperplasia comprising administering an inhibitor selected from a β-arrestin1/2 inhibitor and a GRK2 inhibitor to a subject suffering from at least one of acute or chronic pain, chronic hemolysis (aplastic crises), avascular necrosis, organ damage, and erythroid hyperplasia.
-31. (canceled)
A method of determining the severity of sickle cell disease comprising obtaining a blood sample including red blood cells from a subject;
- optionally treating the red blood cells with at least one of cholera toxin, pertussis toxin or epinephrine;
- and assessing at least one of ERK phosphorylation, GRK2 membrane translocation, or β-arrestin1/2 membrane translocation, wherein the level of ERK phosphorylation, GRK2 membrane translocation, or β-arrestin1/2 membrane translocation is related to the severity of sickle cells disease and/or the likelihood of the red blood cells to adhere to other cells.
