A genetically modified fungal cell of family Hypocreaceae capable to express regulatory protein VEL1 in a higher amount compared to an unmodified wild type fungal cell, comprising:
- a naturally occurring gene encoding the VEL1 protein operably linked to a promoter capable to induce increased expression of the VEL1 protein compared to an unmodified wild type fungal cell;
- or
- additional copies of the naturally occurring gene encoding the VEL1 protein present in the genome of the genetically modified fungal cell.
A vector comprising a nucleic acid molecule encoding a regulatory protein VEL1 of the family Hypocreaceae and having at least 80% identity to SEQ ID No. 1 and a promoter operably linked to the nucleic acid molecule.
A method for producing a recombinant protein in a fungal cell of family Hypocreaceae, comprising:
- providing a wild type fungal cell of family Hypocreaceae;
- genetically modifying the wild type fungal cell by replacing a promoter of the gene encoding the regulatory protein VEL1 by a promoter capable to express the regulatory protein VEL1 in a higher amount compared to the unmodified wild type fungal cell or introducing additional copies of the VEL1 gene into the cell's genome or via a vector;
- introducing into the genetically modified fungal cell a nucleic acid molecule comprising at least one promoter of a cellulase gene, hemicellulase gene, carbohydrate esterase gene and/or gene of other homo- and heteropolysaccharide degrading enzyme operably linked to a heterologous coding region encoding the recombinant protein;
- cultivating the genetically modified fungal cell of family Hypocreaceae in a cultivation medium;
- and
- isolating the recombinant protein produced by the fungal cell.
