Abstract
The present invention relates to systems and methods for manipulating droplets within a high through put microfluidic system.
Claims27
An apparatus for creating a library of emulsion outlooks via a microfluidic system, comprising:
- a carrier fluid channel having a carrier fluid flowing therein at an adjustable or predetermined flow rate;
- a plurality of droplet generators or a plurality of droplet dispensers
- wherein each droplet generator having a reagent fluid supply means for providing reagent fluid to the droplet generator and an outlet channel, the outlet channel in fluid communication with a first fluid channel and a means for transferring a predetermined amount of reagent fluid from the supply means to the outlet channel such that a reagent droplet of predetermined amount is caused to enter the carrier fluid or
- wherein each droplet dispenser having a supply of droplets in the form of a water-in-oil emulsion and an outlet channel, the outlet channel in fluid communication with a first fluid channel and a means for transferring a predetermined volume of emulsion from the supply means to the outlet channel such that a predetermined volume of emulsion is caused to enter the carrier fluid;
- a controller in communication with the plurality of droplet generators for controlling the means for transferring a predetermined amount of fluid in each droplet generator.
2 more elements
A method of creating a library of emulsion outlooks, comprising:
- flowing a carrier oil in a carrier fluid channel;
- releasing a plurality of reagent droplets into the carrier oil;
- maintaining the plurality of reagent droplets in a microfluidic channel;
- flowing a series of emulsion droplets in a micro-fluidic cross-channel in fluid communication with the microfluidic channel;
- introducing the plurality of reagent drops in the predetermined grouping into the micro-fluidic cross-channel;
- and
- causing the plurality of reagent drops in the predetermined grouping to merge with a predetermined one of the emulsion droplets.
