Abstract
The present invention is based in part on a chemically defined method of generating neural stem cells (NSCs) and dopaminergic (DA) neurons from human pluripotent stem cells (hPSCs). The DA neurons of the invention can be derived from hPSCs and NSCs. The present invention also provides reagents and kits useful for the derivation of neural stem cells and dopaminergic neurons from human pluripotent stem cells.
Claims29
A method of producing neural stem cells (NSCs) comprising:
- (a) treating human pluripotent stem cells (hPSCs) with a checkpoint kinase 1 (CK1) inhibitor and a bone morphogenic protein (BMP) inhibitor;
- and
- (b) analyzing the cells for neural stem cell markers.
A method of generating dopaminergic neurons from human pluripotent stem cells comprising:
- (a) treating human pluripotent stem cells (hPSCs) with a checkpoint kinase 1 (CK1) inhibitor and a bone morphogenic protein (BMP) inhibitor;
- (b) identifying neural stem cells by identifying neural stem cell markers;
- (c) treating NSCs with at least one dopaminergic neuron inducing compound;
- and
- (d) analyzing cells for dopaminergic neuron cell markers.
The dopaminergic neuron 25, wherein the dopaminergic neuron inducing compound is Guggulsterone.
A kit for the production of NSCs comprising:
