Abstract
Aspects of the invention relate to novel methods and compositions for assessing the level of cellular uptake of a nanoparticle construct, assessing the level of target binding of a nanoparticle construct and assessing the levels of RNAs and proteins in a given cell.
Claims31
A method for detecting cellular uptake of a nanoparticle construct, comprising:
- providing a nanoparticle construct, comprising:
- a nanoparticle core,
- a first modality comprising a binding moiety specific for a target molecule, that is attached to the nanoparticle core;
- and
- a second modality comprising an uptake control moiety, that is attached to the nanoparticle core and that includes a reference chromophore;
- contacting the nanoparticle construct with a cell;
- and
- detecting the level of the reference chromophore within the cell, wherein the level of the reference chromophore within the cell indicates the level of cellular uptake of the nanoparticle construct within the cell.
