Abstract
Diffuse fluorescence flow cytometers and methods of using them include a plurality of excitation sources and a plurality of detectors, all circumferentially arranged about a space for accommodating a limb of a subject. Tomographic reconstructions of cells within the limb are made by varying the intensity and direction of excitation and then analyzing the results.
Claims11
A diffuse fluorescence flow cytometer, comprising:
- a plurality of excitation sources positioned circumferentially about a space sized for accommodating a limb of a subject, the limb comprising fluorophore-labeled cells circulating therewithin;
- a plurality of detectors positioned circumferentially about the space for collecting a fluorescent emission from the cell;
- and
- a processor responsive to the plurality of excitation sources and the plurality of detectors and configured to generate, based on the states of the excitation sources and of the detectors, a tomographic reconstruction showing the position of the fluorescent emission within of a cross-section of the limb.

A method of determining a position of a fluorophore-labeled cell within a limb of a subject, the method comprising:
- surrounding the limb of the subject with a plurality of excitation sources and a plurality of detectors;
- operating the excitation sources to excite the fluorphore and operating the detectors to detect the fluorophore within the limb;
- and
- based on detections of the fluorophore by the detectors, generating a tomographic reconstruction showing the position of the fluorescent emission within a cross-section of the limb.
