Abstract
Proteins that bind IL-1α and IL-1β are described along with their use in compositions and methods for treating, preventing, and diagnosing IL-1-related disorders and for detecting IL-1α and IL-1β in cells, tissues, samples, and compositions.
Claims26
An isolated composition comprising an anti-IL-1α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, wherein, when administered intravenously to a subject at a dose of about 5 mg/kg, the anti-IL-1α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, exhibits:
- (a) an area under the curve (AUC) of between about 15 and 25 mgh/ml;
- (b) a volume of distribution of between about 85 and 105 mL/kg;
- (c) a half-life of between about 7 and about 13 days;
- or
- (d) a clearance rate of between about 0.1 and about 0.4 ml/h/kg.
An isolated composition comprising an anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, wherein, when administered intravenously to a subject at a dose of about 4 mg/kg, the anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, exhibits:
- (a) an area under the curve (AUC) of between about 10 and 20 mgh/ml;
- (b) a volume of distribution of between about 75 and 95 mL/kg;
- (c) a half-life of between about 7 and about 13 days;
- or
- (d) a clearance rate of between about 0.1 and about 0.4 ml/h/kg.
An isolated composition comprising an anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, wherein, when administered intravenously to a subject at a dose of about 5 mg/kg, the anti-IL-1α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, exhibits:
- (a) an area under the curve (AUC) of between about 15 and 30 mgh/ml;
- (b) a volume of distribution of between about 45 and 75 mL/kg;
- (c) a half-life of between about 7 and about 13 days;
- or
- (d) a clearance rate of between about 0.1 and about 0.4 ml/h/kg.
An isolated composition comprising an anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, wherein, when administered subcutaneously to a subject at a dose of about 5 mg/kg, the anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, exhibits:
- (a) an area under the curve (AUC) of between about 15 and about 30 mgh/ml;
- (b) a half-life of between about 10 and about 30 days;
- or
- (c) a peak concentration (Cmax) of between about 20 and about 40 μg/ml.
An isolated composition comprising an anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, wherein, when administered subcutaneously to a subject at a dose of about 4 mg/kg, the anti-IL-α/β dual variable domain immunoglobulin, or antigen-binding portion thereof, exhibits:
- (a) an area under the curve (AUC) of between about 3 and about 12 mgh/ml;
- (b) a half-life of between about 7 and about 20 days;
- or
- (c) a peak concentration (Cmax) of between about 10 and about 30 μg/ml.
- (a) an area under the curve (AUC) of between about 15 and about 30 mgh/ml;
- (b) a half-life of between about 4 and about 15 days;
