Histone Deacetylase (HDAC) Inhibitors for the Treatment of Cancer

A method of preventing, inhibiting, or arresting cell proliferation of a neoplastic cell, wherein the method comprises contacting the neoplastic cell with a first amount of a histone deacetylase inhibitor and a second amount of an other chemotherapeutic agent, wherein the first and second amounts together comprise an effective amount;

• wherein the histone deacetylase inhibitor is selected from compounds of the following formula and pharmaceutically acceptable salts and solvates thereof:
• wherein:
• A is an unsubstituted phenyl group;
• Q1 is a covalent bond, a C1-7alkylene group, or a C2-7alkenylene group;
• J is:
• R1 is hydrogen, C1-7alkyl, C3-20heterocyclyl, C5-20aryl, or C5-20aryl, C1-7alkyl;
• and,
• Q2 is:
• and
• wherein the other chemotherapeutic agent is selected from:
• an antibody against VEGF, Avastin®, an antibody against CD20, rituximab, bortezomib, thalidomide, dexamethasone, vincristine, doxorubicin, and melphalan.

A kit or kit-of-parts comprising:

• (a) a histone deacetylase inhibitor, preferably as a component of a pharmaceutically acceptable formulation, and preferably provided in a suitable container and/or with suitable packaging;
• and
• (b) another chemotherapeutic agent, preferably as a component of a pharmaceutically acceptable formulation, and preferably provided in a suitable container and/or with suitable packaging;
• wherein the kit or kit-of-parts is suitable for use in a method of treating cancer;
• wherein the histone deacetylase inhibitor is selected from compounds of the following formula and pharmaceutically acceptable salts and solvates thereof:
• wherein:
• A is an unsubstituted phenyl group;
• Q1 is a covalent bond, a C1-7alkylene group, or a C2-7alkenylene group;
• J is:
• R1 is hydrogen, C1-7alkyl, C3-20heterocyclyl, C5-20aryl, or C5-20aryl-C1-7alkyl;
• and,
• Q2 is:
• and
• wherein the other chemotherapeutic agent is selected from:
• an antibody against VEGF, Avastin®, an antibody against CD2O, rituximab, bortezomib, thalidomide, dexamethasone, vincristine, doxorubicin, and melphalan.

A method of treating a hematological cancer comprising administering to a patient in need thereof a therapeutically effective amount of a histone deacetylase inhibitor, wherein the histone deacetylase inhibitor is selected from compounds of the following formula and pharmaceutically acceptable salts and solvates thereof:

• Q1 is a covalent bond, a C1-7alkylene group, or a C2-7alkenylene group;
• J is:
• R1 is hydrogen, C1-7alkyl, C3-20heterocyclyl, C5-20aryl, or C5-20aryl-C1-7alkyl;
• and,
• Q2 is:

A method of preventing, inhibiting, or arresting cell proliferation of a neoplastic cell, wherein the neoplastic cell is a myeloma cell, a plasmacytoma cell, or a plasma cell leukemia cell, wherein the method comprises contacting the neoplastic cell with an effective amount of a histone deacetylase inhibitor,

• wherein the histone deacetylase inhibitor is selected from compounds of the following formula and pharmaceutically acceptable salts and solvates thereof:
