Abstract
The present invention provides methods and uses of neural cells differentiated from adult stem cells of the oral mucosa for cell therapy of neurological and psychiatric diseases and disorders. Methods for direction of differentiation of oral mucosal stem cells into neuronal or neuron supporting cells are also provided.
Claims21
1-42. (canceled)
A method of generating neural cells useful for treating neurological or psychiatric disease or disorder, the method comprising:
- (a) obtaining oral mucosa stem cells (OMSC) from oral mucosa;
- (b) incubating the OMSC of (a) in a pre-differentiating medium, comprising at least one agent selected from the group consisting of:
- N-2 supplement, basic fibroblast growth factor 2 (bFGF) and epidermal growth factor (EGF), capable of inducing the differentiation of OMSC toward a specific neural lineage;
- and
- (c) incubating the OMSC of (b) in a differentiating medium comprising at least one agent selected from the group consisting of:
- dibutyryl cyclic adenosine monophosphate (dbcAMP), IBMX (3-Isobutyl-1-methylxanthine), neuregulin, and platelet-derived growth factor (PDGF), thereby generating cells useful for treating neurological or psychiatric disease or disorder.
3 more elements
A method for differentiating hOMSC into dopaminergic neural cells comprising the steps of:
- (a) incubating hOMSC for at least 48 hours in a pre-differentiation medium;
- and
- (b) incubating the hOMSC of (a) in a differentiation medium for at least 11 days.
