Abstract
A method for quantitatively measuring white blood cell (leukocyte) count and/or white blood cell (leukocyte) subsets count involves adding specific antibodies labeled with a marker to the biological fluid sample, capture of white blood cells from a fluid sample by a retainer, removal of other than leukocyte cells and other interfering substances by washing or using specific magnetic beads, and reading the result. The device for use in the present method includes a retainer for leukocyte cells and an absorption pad for taking up all excess washing solution flowing past the sample.
Claims22
A method for determining of total leukocyte or at least one leukocyte subset in a biological fluid sample, the method comprising the steps of:
- (a) adding to said biological fluid sample of an antibody specific to said total leukocyte or to said at least one leukocyte subset, said antibody conjugated with a marker;
- (b) incubating said biological fluid sample for duration from about 5 min to about 60 min and at a temperature ranging from about 20 degrees Celsius to about 37 degrees Celsius;
- (c) filtering said biological fluid sample through a retainer filter to bind onto said retainer filter of said leukocytes of said biological fluid sample;
- (d) washing said retainer filter to remove weakly-bound substances;
- and
- (e) reading said marker for determining of said total leukocyte or said at least one leukocyte subset in said biological fluid.
A method for determining of total leukocyte or at least one leukocyte subset in a biological fluid sample, the method comprising the steps of:
- (a) adding to said biological fluid sample of an antibody specific to said total leukocyte or to said at least one leukocyte subset, said antibody conjugated with a marker;
- (b) adding to said biological fluid sample of magnetic beads carrying at least one antibody selected to bind to at least one interfering substance;
- (c) removing of said magnetic beads together with said at least one interfering substance from said biological fluid sample using a magnet;
- (d) filtering said biological fluid sample through a retainer filter to bind onto said retainer filter of said leukocytes of said biological fluid sample;
- (e) washing said retainer filter to remove weakly-bound substances;
- and
- (f) reading said marker for determining of said total leukocyte or said at least one leukocyte subset in said biological fluid.
A flow-through cartridge for determining of total leukocyte or at least one leukocyte subset in a biological fluid sample, the cartridge comprising a cartridge housing supporting a fluid sample acceptance tube, an absorbent pad, a removable strip containing a retainer filter configured for binding thereto of said leukocytes of said biological fluid sample, and a disengaging mechanism;
- said retaining filter is placed therebetween and in fluid communication with said fluid sample acceptance tube and said absorbent pad;
- said disengaging mechanism configured to lift said fluid sample acceptance tube from said retaining filter to release thereof for subsequent removal from said cartridge housing using said removable strip.
