Abstract
The present invention provides compositions and methods for colorimetric detection schemes for detecting a variety of biomolecules. The compositions and methods employ DNA hybridization chain reaction for catalytic aggregation of gold nanoparticles. In this catalytic aggregation scheme, a single target DNA strand triggers the formation of multiple inter-particle linkages in contrast to the single linkage formed in conventional direct aggregation schemes.
Claims32
A method for detecting the presence of a target sequence comprising:
- contacting a sample suspected of containing said target sequence, with a plurality of linker sequences that can bind with said target, and with gold nanoparticles, wherein said linkers aggregate in the presence said target sequence;
- and thereafter detecting the target sequence in the sample by colorimetric detection.
A kit comprising:
- gold nanoparticles, linker that binds to a target sequence, and optionally, at least one biological sample preservative or additive, an RNA extractant buffer, a reaction buffer, a negative control sample, a positive control sample, a reaction container, a colorimetric chart, a packaging material, an instruction for use in detecting a said target sequence.
A gold nanoparticle having a spacer group and oligonucleotide linker which will hybridize to a contiguous target sequence a′ and b′, said linker comprising:
- the complement of target sequence a′,
- the complement of target sequence b′;
- a hairpin loop of sequence c;
- and
- the b′ sequence so that b and b′ sequences hybridize to form a hairpin loop.
A gold nanoparticle having a spacer group and oligonucleotide linker which will hybridize to a contiguous target sequence a′ and b′, said linker comprising:
- the complement of target sequence b′;
- a less than full length complement of target sequence a′;
- the complement of hairpin sequence c.
A gold nanoparticle having a spacer group and one of two oligonucleotide linkers which will hybridize with contiguous target sequence a′ and b′, said linker comprising:
- the complement of a′ or the complement of b′.
A method for detecting the presence of a target sequence comprising:
- contacting a sample suspected of containing said target sequence, with first and second linker sequences that bind with said target, and with gold nanoparticles;
- wherein said target comprises sequences a′ and b′ and said first linker includes the complement of a′ and b′ and the sequence b′ with a hairpin, c, and said second linker includes the complement of hairpin c, the complement of b′ and a less than full-length complement of a′ so that said linkers form linker target duplexes and aggregate in the presence said target sequence;
- and thereafter
- detecting the presence of aggregates and thus the target sequence by colorimetric detection.
A method for detecting the presence of a target sequence comprising:
- contacting a sample suspected of containing said target sequence, with first and second linker sequences that bind with said target, and with gold nanoparticles;
- wherein said target comprises contiguous sequences, a′ and b′, said first linker includes the complement of a′;
- and said second linker includes the complement of b′;
- so that said linkers form linker target duplexes and aggregate in the presence of free target sequence, and detecting the presence of aggregates and thus said target sequence by colorimetric detection.
