An antibody composition comprising at least one antibody selected from (a), (b) and (c), wherein the antibody has complex-type and/or hybrid-type N-glycoside-linked sugar chain in a Fc region:
- (a) the antibody has two N-glycoside-linked sugar chains in which fucose is not bound (F0), (b) the antibody has one of N-glycoside-linked sugar chain in which fucose is not bound and one of the complex N-glycoside-linked sugar chain in which fucose is bound (F1), and (c) the antibody has two N-glycoside-linked sugar chain in which fucose is bound (F2);
- wherein the proportion of antibody (a) is greater than the proportion of antibody (b) and antibody (c), that is F0>F1+F2.
A method of producing an antibody composition comprising IgG antibodies, said method comprising increasing a ratio of an antibody (a) compared to an antibody (b) more than 1.0, wherein the antibody has complex-type and/or hybrid-type N-glycoside-linked sugar chains in a Fc region:
- (a) the antibody has two N-glycoside-linked sugar chains in which fucose is not bound, and (b) the antibody has two N-glycoside-linked sugar chains in which fucose is bound;
- and measuring antibody dependent cellular cytotoxicity (ADCC) activity of said composition.
A method of producing an α-1,6-fucosyltransferase knock-out animal cell, said method comprising culturing a cell:
- (i) having a DNA encoding an antibody, and
- (ii) having a lower or no α1,6-fucosyltransferase activity for adding fucose to N-acetylglucosamine of the reducing terminus of N-glycoside-linked sugar chains, by deleting the gene encoding the α1,6-fucosyltransferase or by introducing a mutation to said gene to reduce or eliminate the α1,6-fucosyltransferase activity compared to the cell without said deletion or mutation, wherein said sugar chains comprise
A method of producing an α-1,6-fucosyltransferase knock-out cell, said method comprising culturing an animal host cell having a lower or no α1,6-fucosyltransferase activity for adding fucose to N-acetylglucosamine of the reducing terminus of N-glycoside-linked sugar chains, by deleting the gene encoding the α1,6-fucosyltransferase gene or by adding a mutation to said gene to reduce or eliminate the α1,6-fucosyltransferase activity compared to the cell without said deletion or mutation, wherein said sugar chains comprise
An animal cell:
- (i) carrying a DNA encoding an antibody, and
- (ii) having a lower or no α1,6-fucosyltransferase activity for adding fucose to N-acetylglucosamine of the reducing terminus of N-glycoside-linked sugar chains, by deleting the gene encoding the α1,6-fucosyltransferase or by introducing a mutation into said gene to reduce or eliminate the α1,6-fucosyltransferase activity compared to the cell without said deletion or mutation, wherein said sugar chains comprise
An animal host cell having a lower or no α1,6-fucosyltransferase activity for adding fucose to N-acetylglucosamine of the reducing terminus of N-glycoside-linked sugar chains, by deleting the gene encoding the α1,6-fucosyltransferase gene or by introducing a mutation into said gene to reduce or eliminate the α1,6-fucosyltransferase activity compared to the cell without said deletion or mutation, wherein said sugar chains comprise
