Abstract
The methods of the present disclosure are directed toward methods of diagnosis and treatment of cancer in a subject. The present disclosure provides methods of diagnosing abnormal cells in a subject by measuring the level of Hexim-1 in a sample from the subject, where elevated levels of Hexim-1 indicate abnormal cells. This disclosure further provides methods of treating cancer or hyperplasia, comprising administering to a subject in need thereof a therapeutically effective amount of Hexim-1 or an activator of Hexim-1 expression or activity.
Claims25
A method of detecting abnormal cells in a sample, which comprises measuring the level of Hexim-1 in a cell sample obtained from a subject, wherein abnormal cells are present when elevated levels of Hexim-1 are identified in said sample.
A method of detecting cancer or hyperplasia in a subject, which comprises measuring Hexim-1 levels in the nucleus and cytoplasm of a cell sample obtained from the subject, wherein hyperplasia is identified in said subject by elevated levels of Hexim-1 in the nucleus but not the cytoplasm, and cancer is identified in said subject by elevated levels of Hexim-1 in at least the cytoplasm.
A kit for detecting abnormal cells in a sample from a subject, comprising one or more agents for detection of Hexim-1.
A method of treating cancer or hyperplasia, which comprises administering to a subject in need thereof a therapeutically effective amount of Hexim-1, an activator of Hexim-1 expression or activity, or an inhibitor of Hexim-1 phosphorylation.
A method to detecting improvement or worsening of hyperplasia or cancer in a subject, which comprises measuring the level of Hexim-1 in a cell sample obtained from the subject, wherein improvement of said subject's hyperplasia or cancer is identified by reduced levels of Hexim-1 in said sample, and worsening of said subject's hyperplasia or cancer is identified by increased levels of Hexim-1 in said sample.
A method to determine the effectiveness of a cancer therapy in a subject, which comprises measuring the level of Hexim-1 in a cell sample obtained from the subject, wherein effective cancer therapy is identified by reduced levels of Hexim-1 in said sample, and wherein ineffective cancer therapy is identified by increased levels of Hexim-1 in said sample.
A method of screening for an activator of Hexim-1 expression, the method comprising the steps of (i) providing a cell comprising a Hexim-1 promoter operably linked to a nucleic acid segment encoding an expressible marker;
- (ii) contacting said cell with a candidate substance;
- and (iii) assessing the expression of said marker, wherein an increase in expression of said marker, as compared to expression in a cell not contacted with said candidate substance, identifies said candidate compound as an activator of Hexim-1 expression.
