Abstract
The invention provides silicate prodrugs comprising a therapeutic agent linked to one or more groups of formula (I): —Si(OR)3 (I); wherein each R independently has any of the values defined herein, as well as nanoparticles comprising such compounds.
Claims29
A compound comprising a therapeutic agent linked to one or more groups of formula (I):
- —Si(OR)3 (I)
- wherein:
- each R is indepently selected from (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, wherein each (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, is optionally substituted with one or more hydroxy, (C1-C6)alkoxy, oxo, halo, or aryl;
- or a salt thereof.
The compound of claim 43 wherein an oxygen of a hydroxy group of the taxane is linked to the group of formula (I).
The compound of claim 43 wherein oxygens of at least two hydroxy groups of the taxane are linked to groups of formula (I).
(canceled)
The compound of claim 43 wherein the taxane is a paclitaxel or docetaxel.

A method to treat cancer in an animal comprising administering to the animal a compound as described in claim 43.
29-32. (canceled)
A method to treat restenosis in an animal comprising administering to the animal a compound as described in claim 43.
34-37. (canceled)
A method to treat cancer in an animal, comprising administering to the animal a nano-particle as described in claim 45.
39-42. (canceled)
A compound comprising paclitaxel linked to one or more groups of formula (I):
- —Si(OR)3 (I)
- wherein:
- each R is indepently selected from (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, wherein each (C1-C20)alkyl, (C2-C20)alkenyl, (C2-C20)alkynyl, (C1-C20)alkanoyl, (C2-C20)alkenylcarbonyl, and (C2-C20)alkynylcarbonyl, is optionally substituted with one or more hydroxy, (C1-C6)alkoxy, oxo, halo, or aryl;
- or a salt thereof.
