Abstract
The present invention relates to immunoglobulins that bind IgE and FcγRIIb with high affinity, said compositions being capable of inhibiting cells that express membrane-anchored IgE. Such compositions are useful for treating IgE-mediated disorders, including allergies and asthma.
Claims25
A method of rapidly lowering the serum concentration of an antigen in a patient comprising:
- a) administering an antibody comprising:
- i) a variable region that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein said antibody binds to said antigen to form an antibody-antigen complex and said complex is cleared at least two fold faster than the antigen alone.
A method of lowering the free antigen in a patient comprising:
- a) administering an antibody comprising:
- i) a variable region that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein the concentration of said total antigen rapidly decreases.
A method of differentially clearing an antibody-antigen complex in a patient compared to antibody alone, comprising:
- a) administering an antibody comprising:
- i) a variable region that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein said antibody binds to said antigen to form an antibody-antigen complex and said complex is cleared at least two fold faster than the antibody alone.
A method of rapidly lowering the serum concentration of an antigen in a patient comprising:
- a) administering an Fc fusion protein comprising:
- i) a binding moiety that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein said Fc fusion protein binds to said antigen to form a protein-antigen complex and said complex is cleared at least two fold faster than the antigen alone.
A method of lowering the free antigen in a patient comprising:
- a) administering an Fc fusion protein comprising:
- i) a binding moiety that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein the concentration of said total antigen rapidly decreases.
A method of differentially clearing an antibody-antigen complex in a patient compared to antibody alone, comprising:
- a) administering an Fc fusion protein comprising:
- i) a binding moiety that binds said antigen;
- and
- ii) a variant Fc domain comprising an amino acid substitution as compared to a parent Fc domain wherein said variant Fc domain binds FcγRIIb with increased affinity as compared to said parent Fc domain;
- wherein said Fc fusion protein binds to said antigen to form a protein-antigen complex and said complex is cleared at least two fold faster than the protein alone.
