Abstract
An isolated nucleic acid molecule that encodes a terpene synthase and is selected from among: a) a nucleic acid molecule comprising the sequence of nucleotides set forth in SEQ ID NO: 1, SEQ ID NO: 3 or SEQ ID NO: 5; b) a nucleic acid molecule that is a fragment of (a); c) a nucleic acid molecule comprising a sequence of nucleotides that is complementary to (a) or (b); and d) a nucleic acid molecule that encodes a terpene synthase having at least or at least about 60%, 65%, 70%, 75%, 80%, 85%, 90%, 95%, 96%, 97%, 98%, 99% identity to any one of (a)-(c); wherein the nucleic acid molecule encodes a terpene synthase.
Claims39
An isolated nucleic acid molecule that encodes a Santalum species terpene synthase that comprises the sequence of amino acid residues set forth in one of SEQ ID NOS:2, 4 and 6, wherein the nucleic acid molecule is cDNA.
An isolated terpene synthase, comprising one or more amino acid substitutions in the sequence of amino acid residues set forth in any of SEQ ID NOS:
- 2, 4 or 6, wherein:
- the one or more amino acid substitutions are selected from among substitution of one or more aspartic acid residues with glutamic acid;
- substitution of one or more isoleucine residues with leucine;
- substitution of one or more glycine or valine residues with alanine;
- substitution of one or more tyrosine or phenylalanine residues with tryptophan;
- and/or substitution of one or more proline residues with serine.
An isolated nucleic acid molecule that encodes a terpene synthase from S. spicatum or S. austrocaledonicum, wherein:
- the synthase catalyzes production of one or more of an α-santalene, α-trans-bergamotene, epi-β-santalene and β-santalene;
- and
- the nucleic acid molecule is cDNA.
An isolated terpene synthase comprising the sequence of amino acid residues set forth in SEQ ID NOS:
- 2, 4 or 6.
A host cell, comprising nucleic acid that encodes a Santalum species terpene synthase, wherein:
- the nucleic acid is heterologous to the host cell;
- the encoded synthase catalyzes the production of one or more of α-santalene, α-trans-bergamotene, epi-β-santalene and β-santalene;
- and
- the encoded terpene synthase comprises the sequence of amino acid residues set forth in SEQ ID NOS:
- 2, 4 or 6, or a catalytically active fragment thereof.
A method for detecting the presence of a terpene synthase that catalyzes production of a santalene in a sample, comprising:
- a) contacting a sample suspected of containing the terpene synthase with an antibody that specifically binds to a terpene synthase from a Santalum species under conditions whereby reaction complexes comprising the antibody and the terpene synthase in the sample form;
- and
- b) detecting the formation of reaction complexes comprising the antibody and the terpene synthase in the sample, wherein detection of the formation of reaction complexes indicates the presence of the terpene synthase in the sample;
- or
- c) detecting nucleic acid encoding the terpene synthase.
