Abstract
Provided herein are methods for detecting and/or confirming the neurogenesis effect of cholesterol and/or oxysterol utilizing adipose-derived stem cells (ADSCs). Further provided are methods of promoting neurogenesis in ADSCs. Also provided are methods for the use of cholesterol and/or oxysterol for the production of a neurologic component that may be incorporated into a nutritional composition for promoting neurogenesis in a pediatric subject.
Claims20
A method for detecting neurogenesis of cholesterol or at least one oxysterol, comprising:
- a) culturing adipose-derived stem cells (ADSCs) in the presence of cholesterol or at least one oxysterol;
- and determining the extent of neurogenesis in the ADSCs cultured in the presence of cholesterol or at least one oxysterol;
- b) culturing ADSCs in the absence of cholesterol or at least one oxysterol and determining the extent of neurogenesis in the ADSCs cultured in the absence of cholesterol or at least one oxysterol;
- and
- c) comparing the extent of neurogenesis in the ADSCs cultured in the presence of cholesterol or at least one oxysterol to the extent of neurogenesis of the ADSCs cultured in the absence of cholesterol or at least one oxysterol, and
- d) determining the extent of neurogenesis by observing a change in cell morphology of the ADSCs,
- wherein an increase in the extent of neurogenesis in the ADSCs cultured in the presence of cholesterol or at least one oxysterol compared to the extent of neurogenesis in the ADSCs cultured in the absence of cholesterol or at least one oxysterol indicates that the candidate compound is a neurogenesis-promoting compound.
A method for the use of cholesterol, at least one oxysterol or combinations thereof for the production of a neurologic compound for promoting neurogenesis in a pediatric subject, comprising:
- providing a nutritional composition comprising a carbohydrate source, a fat source, a protein source, and a neurologic component comprising cholesterol, at least one oxysterol, or combinations thereof to a pediatric subject.
A method for the use of a neurologic component comprising cholesterol, at least one oxysterol or combinations thereof for promoting neurogenesis in a pediatric subject, comprising:
- providing a nutritional composition comprising per 100 kcal:
- from about 6 g to about 22 g of a carbohydrate source,
- from about 1.3 g to about 7.2 g of a fat source,
- from about 1.8 g to about 6.8 g of a protein source,
- from about 1 g to about 200 mg of a neurologic component comprising cholesterol, at least one oxysterol and combinations thereof.
