## Abstract

The present disclosure relates to engineered zinc finger proteins that target genes in plants involved in fatty acid biosynthesis. Methods of using such zinc finger proteins in modulating gene expression, gene inactivation, and targeted gene modification are also provided.

## Claims

### Claim 1

A non-naturally occurring zinc finger protein that modulates expression of at least one endogenous Fatty acid thioesterase (Fat) plant gene.

### Claim 2 (depends on 1)

A fusion protein comprising a zinc finger protein of claim 1 and a functional domain.

### Claim 3 (depends on 2)

The fusion protein of claim 2, wherein the functional domain is a transcriptional regulatory domain or a cleavage domain.

### Claim 4 (depends on 1)

The zinc finger protein or fusion protein of claim 1, wherein the plant gene is contained within a Brassica plant.

### Claim 5 (depends on 4)

The zinc finger protein of claim 4, wherein the zinc finger protein modulates expression of a FatA gene.

### Claim 6 (depends on 4)

The zinc finger protein of claim 4, wherein the zinc finger protein modulates expression of a FatB gene.

### Claim 7 (depends on 1)

A polynucleotide encoding one or more zinc finger proteins of claim 1.

### Claim 8 (depends on 7)

A method of modifying one or more genes involved in fatty acid biosynthesis in a plant cell, the method comprising:
introducing, into the plant cell, one or more expression vectors comprising at least one polynucleotide according to claim 7, under conditions such that the one or more zinc finger proteins are expressed and the one or more Fat genes are modified.

### Claim 9 (depends on 8)

The method of claim 8, wherein the modification comprises modulating expression of at least one Fat gene.

### Claim 10 (depends on 9)

The method of claim 9, wherein expression of at least one Fat gene is activated.

### Claim 11 (depends on 9)

The method of claim 9, wherein expression of at least one Fat gene is repressed.

### Claim 12 (depends on 8)

The method of claim 8, wherein the polynucleotide encodes a zinc finger nuclease and at least one Fat gene is cleaved.

### Claim 13 (depends on 12)

The method of claim 12, further comprising the step of introducing a donor nucleic acid such the donor vector is introduced into the site of cleavage by homologous recombination.

### Claim 14 (depends on 8)

A plant cell comprising at least one Fat gene that has been modified by any of the methods of claim 8.

### Claim 15 (depends on 14)

The plant cell of claim 14, wherein fatty acid content within a seed and fatty acid content in the seed is modified.

### Claim 16 (depends on 14)

A plant comprising at least one cell according to claim 14.

### Claim 17 (depends on 16)

The seed or progeny of a plant according to claim 16.

### Claim 18 (depends on 1)

A plant comprising at least one cell comprising the zinc finger protein of claim 1.
