Abstract
The invention provides methods to inhibit the replication of a flavivirus, methods of inhibiting the guanosine triphosphate (GTP)-binding and guanylyltransferase activity of a flavivirus RNA capping enzyme, and methods of treating a subject infected with a flavivirus. The methods can include contacting a flavivirus with an effective amount of a thioxothiazolidine compound described herein, or a derivative thereof, such as a compound of Formula (I).
Claims18
A method to inhibit the replication of a flavivirus comprising contacting a flavivirus with an effective inhibitory amount of a compound of Formula (II):
- wherein
- n is 1, 2, or 3;
- X is S or O;
- R2 is H, OH, alkyl, alkoxy, amino, nitro, halo, or carboxy;
- R3 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R4 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R5 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R6 is H, OH, alkyl, alkoxy, amino, nitro, halo, or carboxy;
- or a salt or solvate thereof;
- thereby inhibiting the replication of the flavivirus.
A method of inhibiting the guanosine triphosphate (GTP)-binding and guanylyltransferase activity of a flavivirus RNA capping enzyme comprising contacting the RNA capping enzyme of a flavivirus with an effective inhibitory amount of a compound of Formula (II):
- wherein n is 1, 2, or 3;
- X is S or O;
- R2 is H, OH, alkyl, alkoxy, amino, nitro, halo, or carboxy;
- R3 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R4 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R5 is H, OH, alkyl, alkoxy, aryloxy, benzyl, benzyloxy, amino, nitro, halo, or carboxy;
- R6 is H, OH, alkyl, alkoxy, amino, nitro, halo, or carboxy;
- or a salt or solvate thereof
- thereby inhibiting the guanosine triphosphate (GTP)-binding and guanylyltransferase activity of the flavivirus RNA capping enzyme.
