Abstract
The invention provides variants of the Azospirillum irakense CelA β-glucosidase that have improve β-glucosidase activity, particularly improved thermoactivity, compared to the wild type enzyme. The invention further provides related polynucleotides, vectors, host cell, and methods for making and using the variants.
Claims4
A method of producing an alcohol comprising:
- a) providing a fermentable sugar produced by a method of converting a biomass substrate to said fermentable sugar, the method comprising contacting the biomass substrate with a recombinant beta-glucosidase polypeptide variant having beta-glucosidase activity, wherein the recombinant beta-glucosidase polypeptide variant comprises an amino acid sequence that has at least 85% identity to SEQ ID NO:4 and comprises a substitution or deletion of the amino acid residue F211, wherein the amino acid sequence is numbered with reference to the amino acid sequence of SEQ ID NO:4;
- b) contacting the fermentable sugar with a fermenting microorganism to produce the alcohol;
- and
- c) recovering the alcohol.
