Abstract
The present invention is directed to compositions and methods for improving the discordance rate and mapping yield in nucleic acid sequencing reactions.
Claims15
A method of sequencing a target sequence of a nucleic acid molecule, the method comprising:
- (a) providing a surface comprising the nucleic acid molecule, the nucleic acid molecule comprising:
- (i) a first adaptor comprising a first anchor site;
- and (ii) the target sequence;
- (b) applying to the surface an aqueous wash solution comprising an effective amount of an acid, a cationic surfactant, or both an acid and a cationic surfactant;
- (c) hybridizing an anchor to the first anchor site;
- (d) extending the anchor to produce an anchor extension product;
- (e) detecting the extension product, thereby identifying a base of the target sequence;
- and
- (f) repeating steps (b) to (e) until the sequence of the target sequence is determined.
An aqueous wash solution configured for sequencing a nucleic acid molecule that is attached to a surface, the wash solution comprising an acid, a cationic surfactant, or both, wherein the wash solution is effective to detectably reduce discordance or to increase a mappable yield by 0.5 percent or more or both compared with a suitable control.
