## Abstract

The present invention relates to compositions and methods for inhibiting or suppressing undifferentiated or pluripotent stem cell growth and proliferation in a differentiated or differentiating cell population or culture.

## Claims

### Claim 1

A method for reducing the number or percentage of pluripotent stem cells in a population of cells, comprising:
contacting a population of cells comprising at least one pluripotent stem cell with a pluripotent stem cell-selective inhibitor or cytotoxic-agent selected from the group consisting of: caffeic acid, Ivermectin, chelerythrine chloride and combinations thereof, thereby reducing the number or percentage of pluripotent stem cells in the population of cells.

### Claim 2 (depends on 1)

The method of claim 1, wherein the pluripotent stem cell is a primate cell.

### Claim 3 (depends on 2)

The method of claim 2, wherein the primate is a human.

### Claim 4 (depends on 1)

The method of claim any of claim 1, wherein the pluripotent stem cell is an ES cell or an iPS cell.

### Claim at position 5

5-6. (canceled)

### Claim 7 (depends on 1)

The method of claim 1, wherein the population of cells is incubated in a cell culture medium containing at least about 1 μM caffeic acid, at least about 1 μM Ivermectin or at least 5 about μM chelerythrine chloride.

### Claim at position 7

8-9. (canceled)

### Claim 10 (depends on 1)

The method of claim 1, wherein the population of cells further comprises differentiating or differentiated cells.

### Claim 11 (depends on 10)

The method of claim 10, wherein the differentiating or differentiated cells are derived from the pluripotent stem cells.

### Claim 12 (depends on 10)

The method of claim 10, wherein the pluripotent stem cell-selective inhibitor or cytotoxic agent has no effect on the viability or proliferation of the differentiating or differentiated cells.

### Claim at position 11

13-14. (canceled)

### Claim 15 (depends on 10)

The method of claim 10, wherein the pluripotent stem cell-selective inhibitor or cytotoxic agent does not inhibit the differentiation or differentiation potential of the cells in the population.

### Claim at position 13

16-20. (canceled)

### Claim 20 (depends on 10)

The method of claim 10, wherein the differentiating or differentiated cells comprise at least one cell type selected from: definitive endoderm cells, PDX1-negative foregut endoderm cells, PDX1-positive foregut endoderm cells, PDX1-positive pancreatic endoderm cells, endocrine progenitor cells and endocrine precursor cells.

### Claim at position 15

21-26. (canceled)

### Claim 27 (depends on 10)

The method claim 10, further comprising confirming a reduction in the number or percentage of pluripotent stem cells in the population of cells.

### Claim at position 17

28-96. (canceled)

### Claim 97

A composition comprising:
a) a pluripotent stem cell, and at least one cell selected from the group consisting of a differentiating derivative of the pluripotent stem cell and a differentiated derivative of the pluripotent stem cell; and
b) a pluripotent stem cell-selective inhibitor or cytotoxic agent selected from the group consisting of: caffeic acid, Ivermectin, chelerythrine chloride and combinations thereof.

### Claim 98 (depends on 97)

The composition of claim 97, wherein the at least one cell is a primate cell.

### Claim 99 (depends on 98)

The composition of claim 98, wherein the primate is a human.

### Claim 100 (depends on 97)

The composition of claim 97, wherein the pluripotent stem cell is an ES cells or an iPS cell.

### Claim at position 22

101-104. (canceled)

### Claim 105 (depends on 97)

The composition of claim 97, wherein the pluripotent stem cell-selective inhibitor or cytotoxic agent has no effect on the proliferation or viability of the differentiating or differentiated cell.

### Claim at position 24

106-119. (canceled)

### Claim 120 (depends on 97)

The composition of claim 97, wherein the pluripotent stem cell-selective inhibitor or cytotoxic agent does not inhibit the differentiation or differentiation potential of the differentiating or differentiated cells in the population.

### Claim 121

A method for reducing the number or percentage of pluripotent stem cells in a population of cells, comprising:
contacting a population of cells comprising at least one pluripotent stem cell and at least one differentiating or differentiated cell with a pluripotent stem cell-selective inhibitor or cytotoxic agent wherein the selective inhibitor or cytotoxic agent is selected from the group consisting of caffeic acid, ivermectin, chelerythrine chloride and combinations thereof, thereby reducing the number or percentage of pluripotent stem cells in the population of cells.
