Abstract
Described herein are microorganisms that produce methionine and related products from endogenous genes in a transsulfuration pathway, as well as from exogenous genes providing a direct sulfhydrylation pathway. Novel genes that are useful for methionine and SAMe production are disclosed.
Claims43
A microorganism comprising:
- one or more first peptides comprising direct sulfhydrylation activity (EC 2.5.1.49 and 4.2.99.-, wherein the direct sulfhydrylation activity uses as a substrate OSHS or OAHS;
- and
- one or more second peptides comprising transsulfuration activity (EC 2.5.1.48, and 4.4.1.8, wherein the direct sulfhydrylation activity consists of expression from an exogenous nucleic acid sequence.
An isolated E. coli comprising a cystathionine gamma synthase (EC 2.5.1.48) or O-succinyl homoserine sulfhydrylase (EC 4.2.99.-) and a homocysteine synthase (EC 2.5.1.49) expressed from an exogenous nucleic acid sequence.
A method of making methionine, comprising
- culturing an isolated microorganism comprising a recombinant DNA molecule, wherein the recombinant DNA molecule over expresses adenylyl sulfate reductase (EC 1.8.99.2) under conditions sufficient for producing methionine;
- and
- isolating methionine.
An isolated peptide comprising a peptide encoded by a nucleic acid sequence selected from SEQ ID NOS:
- 1, 3, 5, 7, or 9, wherein the peptide comprises homoserine O-succinyltransferase activity.
