Abstract
The present invention provides compositions including siderophore receptor polypeptides and porins from gram negative microbes, and preferably, lipopolysaccharide at a concentration of no greater than about 10.0 endotoxin units per milliliter. The present invention also provides methods of making and methods of using such compositions.

A method for detecting antibody comprising:
- contacting antibody with a preparation to result in a mixture, wherein the preparation comprises at least two siderophore receptor polypeptides isolated from a gram negative microbe and lipopolysaccharide at a concentration of no greater than about 10.0 endotoxin units per milliliter (EU/ml);
- incubating the mixture under conditions to allow the antibody to specifically bind a siderophore receptor polypeptide to form an siderophore receptor polypeptide:antibody complex;
- detecting the siderophore receptor polypeptide:antibody complex, wherein detection of the siderophore receptor polypeptide:antibody complex indicates the presence of antibody that specifically binds a siderophore receptor polypeptide.
A method for detecting antibody comprising:
- contacting antibody with a preparation to result in a mixture, wherein the preparation comprises at least two siderophore receptor polypeptides isolated from a gram negative microbe and lipopolysaccharide;
- incubating the mixture under conditions to allow the antibody to specifically bind a siderophore receptor polypeptide to form an siderophore receptor polypeptide:antibody complex;
- detecting the siderophore receptor polypeptide:antibody complex, wherein detection of the siderophore receptor polypeptide:antibody complex indicates the presence of antibody that specifically binds a siderophore receptor polypeptide;
- wherein the concentration of lipopolysaccharide in the preparation is no greater than the concentration of LPS in a reference composition comprising:
- the at least two siderophore receptor polypeptides isolated from the gram negative microbe and lipopolysaccharide;
- wherein the reference composition is produced by a process comprising:
- providing the gram negative microbe;
- disrupting the gram negative microbe in a buffer;
- solubilizing the disrupted gram negative microbe for greater than about 24 hours in a solution comprising sarcosine to result in solubilized and insoluble cellular material, wherein a ratio of the sarcosine to gram weight of disrupted gram negative microbe is between about 0.8 gram sarcosine per about 4.5 grams of disrupted gram negative microbe and about 1.2 grams sarcosine per about 4.5 grams of disrupted gram negative microbe;
- and
- isolating molecules of the gram negative microbe, wherein the isolated molecules comprise the at least two SRPs and LPS.
A method for detecting antibody comprising:
- providing a preparation comprising siderophore receptor polypeptides obtained from a gram negative microbe and lipopolysaccharide at a concentration of no greater than about 10.0 endotoxin units per milliliter (EU/ml);
- contacting an antibody present in a biological sample with the preparation to result in a mixture;
- incubating the mixture under conditions to allow the antibody to specifically bind a siderophore receptor polypeptide to form a siderophore receptor polypeptide:antibody complex;
- detecting the siderophore receptor polypeptide polypeptide:antibody complex, wherein the presence of antibody that specifically binds a siderophore receptor polypeptide indicates the animal may have a condition caused by a microbe expressing siderophore receptor polypeptides.
A method for identifying an SRP comprising:
- contacting an antibody that specifically binds an SRP with a composition to result in a mixture, wherein the composition comprises a polypeptide isolated from a gram negative microbe, wherein the composition comprises lipopolysaccharide at a concentration of no greater than about 10.0 endotoxin units per milliliter (EU/ml);
- incubating the mixture under conditions to allow the antibody to specifically bind the polypeptide to form a polypeptide:antibody complex;
- and
- identifying the presence of the polypeptide:antibody complex, wherein the presence of the polypeptide:antibody complex indicates the polypeptide bound by the antibody is an SRP.
A method for identifying an SRP comprising:
- contacting an antibody that specifically binds an SRP with a composition to result in a mixture, wherein the composition comprises a polypeptide isolated from a gram negative microbe, wherein the composition comprises lipopolysaccharide;
- incubating the mixture under conditions to allow the antibody to specifically bind the polypeptide to from a polypeptide:antibody complex;
- and
- identifying the presence of the polypeptide:antibody complex, wherein the presence of the polypeptide:antibody complex indicates the polypeptide bound by the antibody is an SRP,
- wherein the lipopolysaccharide is at a concentration of no greater than the concentration of LPS in a reference composition comprising:
- the polypeptide isolated from the gram negative microbe and lipopolysaccharide;
- wherein the reference composition is produced by a process comprising:
- providing the gram negative microbe;
- disrupting the gram negative microbe in a buffer;
- solubilizing the disrupted gram negative microbe for greater than about 24 hours in a solution comprising sarcosine to result in solubilized and insoluble cellular material, wherein a ratio of the sarcosine to gram weight of disrupted gram negative microbe is between about 0.8 gram sarcosine per about 4.5 grams of disrupted gram negative microbe and about 1.2 grams sarcosine per about 4.5 grams of disrupted gram negative microbe;
- and
- isolating molecules of the gram negative microbe, wherein the isolated molecules comprise the polypeptide at least two SRPs and LPS.
