Abstract
A polyacrylate formulation suitable for delivery of drug to through a body surface of an individual. By loading the drug and permeation enhancers at a high concentration into a polyacrylate proadhesive that has inadequate adhesive properties for typical adhesive application on the skin, a formulation with desirable adhesive characteristics and effective therapeutic properties can be made. The proadhesive has higher glass transition temperature than typical pressure sensitive adhesives.

- a reservoir including at least one drug, permeation enhancer and an acrylate polymer, wherein the acrylate polymer has soft monomer component of no more than 60 wt %, 40 wt % or more of hard monomer component at least a portion of which is hard polar functional monomer, and 10 to 35 wt % polar functional monomer component at least a portion of which being hard polar functional monomer, the acrylate polymer constituting 45 wt % to 80 wt % of the reservoir and capable of holding the drug and permeation enhancer combination at a dissolved amount of at least 30 wt %, the acrylate polymer having a Tg of greater than −15° C. if without permeation enhancer and without drug, the reservoir being applicable as a pressure-sensitive-adhesive to the body surface for transdermal delivery.
A device for transdermal administration of at least one drug through a body surface of an individual in need thereof, comprising:
- a reservoir including drug and permeation enhancer dissolved in an acrylate polymer, wherein the acrylate polymer has functional group and constituting 50 wt % to 80 wt % of the reservoir and capable of holding the drug and permeation enhancer combination at a dissolved amount of at least 30 wt %, wherein the reservoir is applicable as a pressure sensitive adhesive to the body surface, the acrylate polymer has a Tg of 0 to −15° C., a creep compliance of below 7×10−5 cm2/dyn, and storage modulus of 8×105 dyn/cm2 or above if without drug and permeation enhancer, wherein the reservoir having dissolved therein the drug and permeation enhancer at least 30 wt % has a Tg of −10 to −40° C., a creep compliance of 1×10−4 cm2/dyn to 6×10−4 cm2/dyn and storage modulus of 1.2×105 dyn/cm2 to 6×105 dyn/cm2, the acrylate polymer including a soft monomer alkyl acrylate component, a hard monomer selected from the group consisting of acrylic acid, methyl acrylate, vinyl acetate, and methyl methacrylate and a functional monomer component including at least one of carboxyl group and hydroxyl group.
A device for transdermal administration of at least one drug through a body surface of an individual in need thereof, comprising:
- a reservoir including at least one drug and one or more permeation enhancers dissolved in an acrylate polymer, wherein the acrylate polymer has at 10 wt % functional monomer component and constituting 40 wt % to 80 wt % of the reservoir and capable of holding the drug and permeation enhancer combination at a dissolved amount of at least 30 wt %, wherein the reservoir maintains appropriate pressure sensitive adhesive properties applicable to the body surface, the acrylate polymer has a Tg of 0 to −15° C., a creep compliance of below 7×10−5 cm2/dyn if without the drug and the permeation enhancer, whereas the reservoir with the drug and permeation enhancer at above 30 wt % has a Tg of −10 to −40° C., a creep compliance of 1×10−4 cm2/dyn to 6×10−4 cm2/dyn and storage modulus of 1×105 dyn/cm2 to 8×105 dyn/cm2, the acrylate polymer including a soft monomer alkyl acrylate with a homopolymer Tg of −80 to −20° C., a hard monomer selected from the group consisting of acrylic acid, methyl acrylate, vinyl acetate, methyl methacrylate, acrylamide, and a functional monomer including at least one of acid group and hydroxyl group.
A method of making a reservoir for transdermal drug delivery, comprising:
- providing an acrylate polymer having at least 10 wt % polar functional monomer and capable of holding a drug and permeation enhancer combination in the acrylate polymer at a dissolved amount of at least 30 wt %, incorporating the drug and permeation enhancer in the acrylate polymer to result in a single phase reservoir so that dissolved drug and permeation enhancer constitute together more than 20 wt % and the acrylate polymer constitutes 45 wt % to 80 wt % in solids of the reservoir, wherein the reservoir is applicable as a pressure sensitive adhesive to the body surface.
A method of making a transdermal drug delivery reservoir, comprising:
- providing for a reservoir a polyacrylate proadhesive containing polar functional groups and if solvent-free having a Tg of greater than −15° C., creep compliance of 6×10−5 cm2/dyn to 2×10−6 cm2/dyn, and storage modulus of 8×105 dyn/cm2 or above, incorporating drug and permeation enhancer in the proadhesive with a dissolved concentration therein of greater than 30 wt % of drug and permeation enhancer combination and removing solvent such that the resulting reservoir has pressure sensitive adhesive property appropriate for transdermal drug delivery, the resulting reservoir having a Tg of −10 to −30° C., a creep compliance of 1×10−4 cm2/dyn to 6×10 cm2/dyn and storage modulus of 1×105 dyn/cm2 to 8×105 dyn/cm2.
