Abstract
Disclosed are embryonic stem cell-derived dendritic cells, genetically modified immature dendritic cells capable of maturation, as well as methods for the production of such cells. In one embodiment, the cells made be produced by a method comprising the steps of providing a population of embryonic stem cells; culturing the embryonic stem cells in the presence of a cytokine or combination of cytokines which brings about differentiation of the embryonic stem cells into dendritic cells; and recovering the dendritic cells from the culture. In a further embodiment, the cells may be genetically modified.

A method of screening a candidate drug for an effect on a cultured dendritic cell, the method comprising:
- providing a cultured es dendritic cell (esDC), wherein the esDC is a dendritic cell that is a obtained by culturing an embryonic stem cell in vitro;
- contacting the cultured esDC with a candidate drug;
- and
- assessing an effect of the candidate drug on the cultured esDC.
A method of assessing the effect of a gene on a cultured dendritic cell, the method comprising:
- providing a genetically modified cultured es dendritic cell (esDC) expressing a heterologous gene, wherein the genetically modified cultured esDC, wherein the esDC is a dendritic cell that is a obtained by culturing an embryonic stem cell in vitro;
- providing a cultured es dendritic cell (esDC), wherein the esDC is a dendritic cell that is a obtained by culturing an embryonic stem cell in vitro, wherein the cultured esDC is not genetically modified to express the heterologous gene;
- and
- assessing an effect of the heterologous gene on the genetically modified cultured dendritic cell by comparing the genetically modified cultured dendritic cell to the cultured dendritic cell.
A first in vitro cell population comprising embryonic stem cells and a second in vitro cell population comprising progeny of a portion of the first in vitro cell population, wherein the embryonic stems cells are human or mouse embryonic stems cells, and wherein the progeny are dendritic cells.
