Abstract
The invention relates to monovalent ligands of the human CD28 receptor, which can selectively block the interaction between CD28 and B7 without activating the CD28 receptor. Said ligands can be used in particular in the production of immunosuppressant drugs, selectively blocking T lymphocyte activation phenomena involving the CD28 receptor.
Claims4
A monovalent human CD28 receptor ligand which selectively blocks CD28/B7 interaction without activating the human CD28 receptor, wherein the ligand comprises the CDR5 (complementary determining regions) of the heavy chain and the CDR5 of the light chain of an anti-CD28 antibody which recognizes an epitope formed by portions of amino acid sequences from SEQ ID NO:
- 1 selected from SEQ ID NO:
- 2;
- SEQ ID NO:
- 3;
- SEQ ID NO:
- 4, wherein the ligand is chosen from:
- (a) the Fv, Fab or scFv fragments of said anti-CD28 antibody;
- and
- (b) recombinant proteins comprising the fragment (a) and a heterologous polypeptide;
- with the exception of a ligand containing the CDR5 of the heavy chain and the CDR5 of the light chain of the CD28.3 immunoglobulin.
