Abstract
Safe and effective live vaccines against bacteria infecting aquatic animals were created through the induction of novobiocin-resistance in liquid culture and novobiocin- and rifampicin-resistance in liquid culture.
Claims34
An isolated attenuated bacterial strain from the genus Edwardsiella wherein said strain is resistant to novobiocin or a novobiocin-like antibiotic and is effective for eliciting an immune response in aquatic animals and is protective against infection by virulent strains of the genus Edwardsiella.
An isolated attenuated Edwardsiella ictaluri strain NRRL B-50348.
An isolated attenuated Edwardsiella tarda strain NRRL B-50467.
An isolated attenuated Edwardsiella tarda strain NRRL B-50468.
An isolated attenuated bacterial strain from the genus Streptococcus wherein said strain is resistant to novobiocin or a novobiocin-like antibiotic and is effective for eliciting an immune response in aquatic animals and is protective against infection by virulent strains of the genus Streptococcus.
An isolated attenuated Streptococcus strain selected from the group consisting of Streptococcus iniae and Streptococcus agalactiae.
An isolated attenuated Streptococcus iniae strain NRRL B-50368.
An isolated attenuated Streptococcus agalactiae strain NRRL B-50460.
A vaccine composition for protecting aquatic animals against infection by bacteria from the genus Edwardsiella comprising:
- a. an immunologically effective amount of an isolated attenuated strain of Edwardsiella resistant to novobiocin or a novobiocin-like antibiotic, and
- b. a carrier.
A vaccine composition for protecting aquatic animals against infection by bacteria from the genus Streptococcus comprising:
- a. an immunologically effective amount of an isolated attenuated strain of Streptococcus resistant to novobiocin or a novobiocin-like antibiotic, and
- b. a carrier.
An isolated attenuated strain of Aeromonas hydrophila resistant to novobiocin and rifampicin and effective for elicting an immune response in aquatic animals which is protective against infection by virulent strains of Aeromonas hydrophila.
A vaccine composition for protecting fish against infection by virulent strains of Aeromonas hydrophila comprising:
- a. an immunologically effective amount of an isolated attenuated novobiocin-resistant and rifampicin-resistant strain of Aeromonas hydrophila, and
- b. a carrier.
A method for making attenuated bacteria for use in a vaccine comprising:
- (a) determining the minimum dose of an antibiotic that allows overnight growth of a bacteria in liquid culture media,
- (b) passaging said bacteria into a liquid culture medium having an increased dose of said antibiotic, and
- (c) repeating step (b) until said bacteria is attenuated.
