A method of producing a population of isolated non-human mammalian pluripotent cells that are substantially free from other cell types, the method comprising the step of culturing the pluripotent cells in the presence of an activin agonist and, optionally, FGF2, wherein the cells
- a) are unresponsive to LIF;
- and
- b) are obtained from the late epiblast layer of a post-implantation non-human mammalian embryo in which the late epiblast layers have been microdissected away from extraembryonic tissues.
