Abstract
The invention concerns a novel method for making herbicide-tolerant plants, in particular to HPPD inhibiting herbicides, the nucleic acid sequences coding for enzymes capable of being used in said method, expression cassettes containing them and transgenic plants comprising at least one of said expression cassettes.
Claims29
An isolated polynucleotide selected from the group consisting of
- a) a polynucleotide that encodes an HPP oxidase which is selected from the group consisting of the amino acid sequence of SEQ ID No 2, SEQ ID No 4, SEQ ID No 6, the fragments thereof and homologous sequences thereto which conserves the function of the reference sequence;
- and polynucleotides capable of selectively hybridizing to said polynucleotide that encodes a protein of SEQ ID No 2, SEQ ID No 4, or SEQ ID No 6 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS;
- b) a polynucleotide that encodes an HPAH which is selected from the group consisting of the amino acid sequence of SEQ ID No 8 and SEQ ID No 18;
- the fragments thereof and homologous sequences thereto which conserves the function of the reference sequence;
- and polynucleotides capable of selectively hybridizing to said polynucleotide that encodes a protein of SEQ ID No 8 and SEQ ID No 18 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS;
- c) a polynucleotide that encodes an HPAC which is selected from the group consisting of the amino acid sequence of SEQ ID No 10, SEQ ID No 12, SEQ ID No 14 and SEQ ID No 20;
- the fragments thereof and homologous sequences thereto which conserves the function of the reference sequence;
- and polynucleotides capable of selectively hybridizing to said polynucleotide that encodes a protein of SEQ ID No 10, SEQ ID No 12, SEQ ID No 14 and SEQ ID No 20 under the following conditions:
- prehybridization for four hours at 68° C. in a solution of 5×SSC, 6% Denhardt's, and 0.5% SDS;
- hybridization for sixteen hours at 68° C.;
- and washes at 65° C. in 2×SSC, 0.1% SDS.
