## Abstract

We disclose a vaccine comprising a pappalysin and vaccine compositions comprising a pappalysin.

## Claims

### Claim 1

A vaccine composition comprising a pappalysin polypeptide, or antigenic part thereof, and an adjuvant and/or carrier.

### Claim 2 (depends on 1)

The composition of claim 1 wherein the pappalysin polypeptide comprises the amino acid sequence of SEQ ID NO. 2.

### Claim 3 (depends on 1)

The composition of claim 1 wherein the pappalysin antigenic part comprises an amino acid sequence selected from the group consisting of SEQ ID NO: 23, SEQ ID NO: 24, SEQ ID NO: 25, SEQ ID NO: 26, SEQ ID NO: 27, SEQ ID NO: 28 and variants thereof, wherein the variant is an amino acid addition, deletion or substitution of at least one amino acid residue and wherein the variant includes at least one antigenic epitope.

### Claim 4 (depends on 3)

The composition of claim 3 wherein the pappalysin antigenic part comprises the amino acid sequence of SEQ ID NO: 25 or a variant thereof, wherein the variant is an amino acid addition, deletion or substitution of at least one amino acid residue and wherein the sequence variant includes at least one antigenic epitope.

### Claim 5

A DNA vaccine composition comprising a nucleic acid molecule encoding a pappalysin polypeptide or antigenic part thereof.

### Claim 6 (depends on 5)

The DNA vaccine of claim 5 wherein the nucleic acid molecule is selected from the group consisting of:
i) SEQ ID NO: 1; and
ii) a nucleic acid molecule the complementary strand of which hybridizes under stringent hybridization conditions to SEQ ID NO: 1, wherein the nucleic acid molecule encodes a pappalysin polypeptide or antigenic part thereof.

### Claim 7 (depends on 6)

The DNA vaccine of claim 6 wherein the nucleic acid molecule is selected from the group consisting of:
i) SEQ ID NO: 11, SEQ ID NO: 12, SEQ ID NO: 13, SEQ ID NO: 14, SEQ ID NO: 15, SEQ ID NO: 16; and
ii) a nucleic acid molecule the complementary strand of which hybridizes under stringent hybridization conditions to SEQ ID NO: 11, SEQ ID NO: 12, SEQ ID NO: 13, SEQ ID NO: 14, SEQ ID NO: 15, or SEQ ID NO: 16, wherein the nucleic acid molecule encodes a pappalysin polypeptide or antigenic part thereof.

### Claim 8 (depends on 7)

The DNA vaccine of claim 7 wherein the nucleic acid molecule comprises SEQ ID NO: 13.

### Claim 9 (depends on 5)

The DNA vaccine of claim 5 wherein the nucleic acid molecule is part of an expression vector adapted to express a pappalysin polypeptide or antigenic part thereof.

### Claim 10 (depends on 1)

The composition of claim 1 wherein the adjuvant is a cytokine selected from the group-consisting of GMCSF (granulocyte colony-stimulating factor), interferon gamma, interferon alpha, interferon beta, interleukin 12, interleukin 23, interleukin 17, interleukin 2, interleukin 1, TGF (transforming growth factor), TNFα (tumor necrosis factor alpha), and TNFβ (tumor necrosis factor beta).

### Claim 11 (depends on 1)

The composition of claim 1 wherein the adjuvant is a TLR (toll-like receptor) agonist.

### Claim 12 (depends on 11)

The composition of claim 11 wherein the TLR agonist is selected from the group consisting of: CpG oligonucleotides, flagellin, monophosphoryl lipid A, poly I:C and derivatives thereof.

### Claim 13 (depends on 12)

The composition of claim 12 wherein the adjuvant is a CpG oligonucleotide.

### Claim 14 (depends on 1)

The composition of claim 1 wherein the adjuvant is a bacterial cell wall derivative selected from the group consisting of muramyl dipeptide (MDP) and trehelose dycorynemycolate (TDM).

### Claim 15

A method of vaccinating a subject suffering from or having a predisposition to cancer comprising administering an effective amount of a vaccine composition comprising a pappalysin polypeptide, or antigenic part thereof and an adjuvant and/or carrier.

### Claim 16 (depends on 15)

The method of claim 15 wherein the cancer is prostate cancer.

### Claim 17 (depends on 15)

The method of claim 15 wherein the cancer is lung cancer.
