Abstract
The present invention provides an approach for the simultaneous determination of the activation states of a plurality of proteins in single cells. This approach permits the rapid detection of heterogeneity in a complex cell population based on activation states, and the identification of cellular subsets that exhibit correlated changes in activation within the cell population. Moreover, this approach allows the correlation of cellular activities or properties. In addition, the use of potentiators of cellular activation allows for characterization of such pathways and cell populations.
Claims45
1-13. (canceled)
A method of classifying an acute myeloid leukemia (AML) patient comprising:
- contacting a cell from a patient suspected of having acute myeloid leukemia with a potentiator;
- identifying a potentiated response of Stat5 or Stat3, wherein said identification is performed in a single cell from a patient having a Flt3 mutation;
- and
- correlating the potentiated response to a cell signaling state identified with acute myeloid leukemia.
A method of classifying an acute myeloid leukemia (AML) cell, comprising:
- providing an AML cell to be analyzed that has been contacted with one or more potentiators;
- and
- identifying, in said single AML cell the state of one or more signaling nodes in response to the one or more potentiators;
- and
- classifying said AML cell based on said identification of said state of one or more signaling nodes.
The method of claim 32 wherein said one or more binding element are antibodies.
A method for analyzing cells from a patient suspected of having acute myeloid leukemia (AML) comprising:
- providing a sample of cells from said patient;
- contacting said cells with a potentiator;
- individually determining the activation state of a plurality of signaling nodes in one or more of said cells;
- and
- correlating the activation state of the plurality of signaling nodes in said one or more cells with the activation states of a plurality of signaling nodes of AML cells which have been contacted with the potentiator.
A method of identifying an acute myeloid leukemia (AML) cell comprising:
- providing flow cytometry data generated from a cell suspected of being an AML cell after exposing said cell to a potentiator;
- identifying an activation state of a signaling node in said flow cytometry data, wherein the signaling node can exist in at least two different activation states;
- and
- correlating the signaling node to a signature identified for an AML cell;
- whereby said cell is identified as an AML cell or as a non-AML cell based on said correlating.
