Abstract
The method of biotechnological production of bovine hemoderivative is presented in the following overview providing a summary of individual technological operations in time sequence (Flow-Sheet).
Claims4
A method of biotechnological production of bovine hemoderivative, the method comprising:
- freshly acquired animal blood mass, specifically fresh bovine blood, is first fermented in several phases, usually in two phases, under increased heating air temperature,
- the obtained blood fermentation product is dried,
- the dried blood fermentation product is desintegrated,
- this is followed by ethanol extraction of the blood fermentation product in several phases,
- the blood fermentation product is subjected to one vacuum inspissation, usually to a resulting concentration that is approx 20 times higher than the original concentration, with subsequent removal of undesirable substances,
- the obtained condensed extract is subjected to one ether preparation in which the condensed blood fermentation product is subjected to ether precipitation, and the obtained precipitate is separated from
- the solution of undesirable substances dissolved in ether,
- finally, standardization of the final product is carried out to a concentration of the bovine hemoderivative in water,
- is characterized in that:
- for fermentation, the immediately acquired animal blood mass, preferably bovine blood, is put to small containers with blood mass level height of 2.5-3 cm, the blood mass is gradually heated till the heated blood reaches the temperature 55-65° C. in its mass, preferably 60° C., and then the second fermentation phase begins in which the temperature of the surrounding heating air is reduced, from a temperature approx. 80° C. in the first fermentation phase, to 65-75° C., preferably 70° C., and when this temperature is reached the second fermentation phase proceeds during which a constant temperature difference of 5° C. is maintained between the heating air and the temperature of the blood mass, during which phase autoenzymatic cleavage of blood takes place till the entire blood mass passes into a gummy consistency of the blood fermentation product,
- the obtained blood fermentation product is dried through controlled ventilation according to a drying curve at 60-80° C. for 120-160 hours, preferably at 70° C. and for 140 hours, to a final humidity content of up to 10%, preferably 2-3%,
- the dried blood fermentation product is subsequently disintegrated to particles sized 1-10 mm, preferably 2-4 mm,
- the separated blood fermentation product is subjected to continual extraction in several phases, preferably in 4 phases, through the action of permanently pure ethanol in an extractor of Soxhlet type, using a total extraction time of 150-400 hours, preferably 240 hours, at 70-78° C., the extracts of blood fermentation products are joined during repeated extraction,
- the joined extracts of blood fermentation product obtained in this way are subjected to stabilization of this fermentation product through separation of undesirable blood salts, at ambient temperature for 24-120 hours, preferably 72 hours, whereby the total stabilization time is counted from the beginning of the process of obtaining the last blood fermentation product extract,
- after subsequent vacuum inspissation, the stabilized condensed blood fermentation product extract is subjected to ether preparation in several phases to obtain a precipitate of the bovine hemoderivative, with each precipitate formed in this way being subjected to dissolution in ethanol in amount till quantitative dissolution of the bovine hemoderivative is attained and a true solution of the bovine hemoderivative for the subsequent ether preparation, and the solution overlying the decanted precipitate, containing undesirable phospholipids soluble in ether, is separated,
- after this repeated preparation, the obtained bovine hemoderivative precipitate is stabilized in order to separate the residual undesirable ether from this precipitate,
- the bovine hemoderivative obtained in this way is first dissolved in ethanol till true solution forms, and this solution is then subjected to vacuum inspissation at 25-40° C., preferably at 28° C., till plain ether distillate is obtained, whereupon standardization of the final product is carried out.
