Abstract
The present invention provides novel methods and compositions for the diagnosis and treatment of colon cancers. In particular, the present invention provides diagnostics and prognostics for colon (including colon adenocarcinoma) cancer patients, wherein the methods related to measuring miR levels can predict poor survival. The invention also provides methods of identifying inhibitors of tumorigenesis.
Claims18
A method of diagnosing whether a subject has a poor survival prognosis colon adenocarcinoma, comprising:
- measuring the level of at least one miR-181b gene product in a test sample from the subject wherein said subject has colon adenocarcinoma,
- wherein an increase in at least the level of the miR-181b gene product in the test sample, relative to the level of a corresponding miR-181b gene product in a control sample, is indicative of the subject having poor survival prognosis colon adenocarcinoma.
A method of testing for poor survival prognosis colon adenocarcinoma, which comprises:
- (1) determining an expression level of at least one marker in a sample from a test subject having colon adenocarcinoma;
- the at least one marker including at least one miR-181b gene product;
- (2) comparing the expression level determined in step (1) with a control expression level of the marker in a sample from a healthy subject;
- and
- (3) judging the subject to have a poor survival prognosis colon adenocarcinoma when the result of the comparison in step (2) indicates that:
- the expression level of the at least one marker in the test subject is higher than that in the control.
A method of diagnosing whether a subject has poor survival prognosis colon adenocarcinoma, comprising:
- (1) reverse transcribing RNA from a test sample obtained from the subject to provide a set of target oligodeoxynucleotides wherein said subject has colon adenocarcinoma;
- (2) hybridizing the target oligodeoxynucleotides to a microarray comprising miRNA-181b specific probe oligonucleotides to provide a hybridization profile for the test sample;
- and
- (3) comparing the test sample hybridization profile to a hybridization profile generated from a control sample,
- wherein an increase in the signal of the miR-181b is indicative of the subject having poor survival prognosis colon adenocarcinoma.
