Abstract
The present invention relates to complexes comprising one or more markers and one or more biomolecules for use as a contrast agent in positron emission tomography based imaging. The complexes according to the present invention preferably accumulate in a target compartment, such as the kidney. The present invention further relates to methods for generating the complexes and methods for using the complexes such as for evaluation of different aspects of kidney functionality e.g. for calculation of total and/or regional glomerular filtration rate in the kidney without the need for sampling blood or urine.

A method for generating an image of a target compartment of an individual, comprising the steps of
- administering to the individual a contrast agent comprising one or more complexes, wherein each complex comprises at least one biomolecule and at least one marker whereby optionally, the biomolecule is coupled to the marker via a linker, wherein at least one marker is a Positron Emission Tomography (PET) isotope and the biomolecule is selected from the group consisting of polypeptides, nucleic acids, polysaccharides and lipids,
- visualizing the one or more complexes of the contrast agent by employing a Positron Emission Tomography (PET) based method, and
- capturing one or more images of said target compartment.
A method for assessment of a target compartment functionality of an individual, comprising the steps of:
- obtaining one or more images of said target compartment by
- administering to the individual a contrast agent comprising one or more complexes, wherein each complex comprises a biomolecule and a marker whereby optionally, the biomolecule is coupled to the marker via a linker, wherein the marker is a Positron Emission Tomography (PET) isotope and the biomolecule is selected from the group consisting of polypeptides, nucleic acids, polysaccharides and lipids,
- visualizing the one or more complexes of the contrast agent by employing a Positron Emission Tomography (PET) based method, and
- capturing said one or more images of said target compartment;
- analysing said one or more images;
- and
- assessing the functionality of said target compartment based on a comparison of the obtained one or more images with a physiological standard image from a healthy normal individual.
A method for the assessment of kidney function in an individual, comprising the steps of:
- a) administering to an individual a complex or a composition comprising a Megalin and/or Cubulin ligand linked to a marker which is a Positron Emission Tomography (PET) isotope,
- b) visualizing the complex or the composition administered in step a) by a Positron Emission Tomography (PET) based method, and capturing one or more images of the kidney of said individual,
- c) administering to the same individual a complex or a composition comprising a Megalin and/or Cubulin ligand different from the one used in step a) linked to a marker,
- d) visualising the complex or composition administered in step c) by the PET based method of step b) and capturing one or more images of the kidney of said individual, and
- e) profiling kidney function in said individual based on a comparison of the obtained one or more images for each complex administered with a physiological standard image from a healthy normal individual for said one or more complexes.
