Abstract
Sequencing methods that use an exonuclease that comprises template dependent nucleobase binding activity are provided. Related compositions and sequencing systems are also provided.

A sequencing system, the system comprising:
- a reaction chamber;
- a detector configured to detect a signal from the reaction chamber, which signal results from a transient template-dependent binding of a labeled nucleobase by an enzyme in the reaction chamber;
- and,
- a sequence assembly module that assembles template nucleic acid sequence information based upon detection of the signal.
